I have to wonder (not going to ask directly...) just really how benign our friendly neighborhood Chemdelic actually is. Just kidding. Heavy chemistry, a relaxing voice, the occasional dark humor...what's not to recommend these videos to the Pure Unintentional ASMR channel for immediate consideration? To be clear, I don't find this boring in any way. Though I lack the background (mostly in every way), I do still enjoy it very much; and knowledge is cumulative, for the most part, after all. If you happen to have any broken glass or lab glassware then you can break that into smaller pieces and use that as boiling chips to prevent pumping to some degree.
I got one question. You showed at 12:11 how the Ehrlich reagent forms a stabilized structure at the C3 position of the indoles. How does this work with LSD-25? There the C3 is already substituted and could not make another bond, without breaking aromaticity, right? So wouldn't it rather form a bond with the Ehrlich at the C2?
I was tought these reactions for almost all of these common and more uncommon reagents in pharmacy school. Many of these mechanisms arent well studied and you can expect very different reaction mechanisms depending on each specific compound. For some popular ones there are reasonable proposed mechanisms (Vitali morin for tropaalkaloids or ehrlich for primary amines, phenols hydrazides and indols which is actually the van urk reaction), for some like the marquis reagent, the general proposed mechanism is the that the formaldehyde attacks activated aromatic rings and forms larger conjugated ring systems with deprotonated phenol groups to make an oxonol dye. This is probably the case with morphine, however it might differ greatly for different drugs. Just as an example.
Ehrlich reagent also was once used to distinguish different amino acids in TLC of protein hydrolysates after a first round of staining with ninhydrin. You could separate all twenty on a 2-d TLC plate by eluting with different solvents, I forget the details of which eluents were used. I seem to recall that amino acids other than tryptophan took up the stain at different colors, although tryptophan was obviously the strongest staining.
Even tho dance safe sells their test kits for none to little profit, I like that if the world ends I find a dealers stash I can test it to make sure my meth, heroin and coke don't have fentanyl in them. That's when all the good pharmaceutical drugs I stole are all gone :)
