      That's why he spends months on it probably. If he wasn't being lazy he could finish most projects within a few days to weeks (excluding the time he actually has to wait for some stuff).

    The soxhlet extractor is one of my favorite pieces of glassware. It's extremely useful, even in the culinary world. I use it with ethanol (ever-clear) and ground dried ghost peppers to extract capsaicin to enhance hot sauces.

    Dear NileRed, pure capsaicin isn't very spicy because it requires a carrier oil to stick to the mucous membranes and cause irritation (aka the burning sensation). Could it be that Piperine requires a carrier oil as well? Couldn't find anything online to confirm this.

      Try olive oil (xv) or black walnut oil. These seem good as they cut white(yellow) phosphorus for demos.

      Probably not, as he could still taste the spiciness. What he meant by "nothing" was probably that it was ONLY the spiciness, usually in food spiciness is just one flavor amongst many, so the burning sensation has background flavors that are actually tasteable, and not just sensations.

    I'm four semesters into my chemistry bachelor and we just started with the organic lab last Monday and boy - my appreciation for your videos has easily quadrupled since then. Organic Chemistry is so tedious and takes so long and you go through all the hard work just to present us the cool stuff condensed into 10-20 minutes.

    It makes sense that it doesn't taste like anything. The piperine only interacts with heat-sensitive membrane proteins and not other proteins related to taste.
    So it's basically just concentrated "heat"

    When doing a soxlhet extraction, I would always put my material in a reagent tube made from fine mesh cotton, and plug it with cotton - this would enable you to powder the pepper corn as fine as you wish, increasing the yield

    Fun-fact: Cosmetics industries uses capsaicin along side with epiderfill on lipsticks in order to cause a minor irritation that make the lips grow in size by just a little bit.
    Also, as a chemist undergraduate, i love your videos and i forbid you to stop making them.

      As the hot solvent recondense on top of the crushed pepper it pass through it and carries the piperine with it
      Hes just using some fancy pants glassware(Soxhlet extractor) to keep things simple and easy.
      You can alternatively extract by drag if i'am not mistaken, but as he stated is much more 'labor intensive'.

      That's pretty much it. The Soxhlet extractor, keeps recycling the same alcohol, so that he ends up with a more concentrated solution at the end, than if he just heated the pepper in a large quantity of alcohol and boiled it off.
      The extract works as a d distillation unit, so the the evaporated alcohol is mostly pure when it condenses at the top. It's hot and as it drains through the pepper, it dissolves the piperine.
      When the siphon tube fills to the top, it quickly drains the solution with the dissolved piperine, and the cycle repeats.
      The siphon tube is working with the same principle as a flush toilet.

    Have you considered dissolving the piperine in some sort of cooking oil and tasting it afterwards? I guess due to the poor solubility there's no chance for the piperine to unleash its full potential on your receptors..

      Full immersion + filtration. I would guess it’d taste like what you’d get from using a clever dripper.
      This gives me an idea for a video *runs to cupboard and has neither a clever or a Soxhlet extractor*.... never mind

      That was my thought too. But reboiling your coffee over and over will make it terrible... perhaps not if you do it at low pressure so it boils cooler.

      That is called a perc pot. Brewing coffee with a perc pot is an art. You have to stop it at the right point or it gets bitter.

      Cheeser100 The isopropanol is a bit toxic, it is an irritant. It should have evaporated off though. My point is that in theory it could hurt you in one or a few ways.

    I love Soxhlets 😄 They're super convenient, awesome to look at while extracting and generally have a super smart design. Only used it once tho (for piperine as well). But I used a cellulose based extraction tube instead of cotton. The solvent passes through the pepper a lot nicer and it was easier to remove the pepper after the extraction was finished.

    This is late but:
    Use a BURR GRINDER for black pepper, or in general, if you want a controlled fineness to your grains. Blade grinders just chop in a mess.

    For the extraction from fine powders you may want to use a soxhlet thimble or capsule. These things leave enough room on the sides to let the solvent drain,while offering the increased specific surface area of fine powders. They are commercially available, but you should be able to make one yourself.

    We did this in organic chemistry last fall! Was really fun to do. While checking the melting point to see how pure it was, the entire lab coughed like crazy, and one of the lab engineers had to step outside to get some air :P

    If you don't have any extractor thimbles, you can use scrunched up coffee filters. Just make sure that the reflux condenser drips inside the top of the filter for best results. As long as the top of the filter is above the top of the siphon, it will work fine and the grind of your powder will not be as critical.

    I remember trying to do this in High School during lunchtimes! We didn't have a soxhlet extractor so we just had to boil it under reflux for a few hours, and we had to order in KOH (which didn't arrive in time), but I did get a few of those crystals

    with very fine powders, acid-washed sea sand can be mixed in with it, this allows the solvent to flow through even very fine powders, which increases the extraction enormously
    finely ground substances naturally increase the yield and the sea sand allows the solvent to penetrate the powder very well
