I created the protocol in the instrument, but I was wondering if there's any possibility to export as an excel document and try to use it in the computer? Hi Rita Corrales For analyzing the data in other computer or laptop with Gen5 software you have to save your experiment data in .xpt format during data acquisition. you can't do this from excel format file. you will not be able to import your excel format (.exl) experiment file in Gen5 software to further analyze your data. sorry for this

Sir, what what is corrected blank. Do i need to analyze corrected blank readings or should i go with blank only? The average is coming negative for all the samples after corrected blank.
Pathlength correction is used for Normalization of Absorbance when we use Cuvette or Biocell for detection of DNA or RNA. In this case many factors (like Liquid volume, Plate type and manufacturer, Buffer composition, Nucleic acid concentration, Temperature of the solution) affect the the absorbance . I suggest you please dont use pathlength correction use BLANK substraction only. In case of background correction using Primary (For Exm. 450nm) and Reference (For Exm. 650nm) wavelength. use data reduction option in Protocol and here in Delta transformation use Data In1: Primary (For Exm. 450nm) Data In 2: Reference (For Exm. 650nm) It substract OD value of Reference from Primary. Hello Sabir ji First open the device manager in your laptop or desktop and click on the down arrow sign \/ at Port (COM & LPT) and you can see COM port like COM2 COM4 or etc now connect the usb cable of Reader Machine and you can see new COM port like USB Serial Port (COM9). Now open the Gen 5 software and click on System and then open Instrument Configuration. In the Intrument configuration click on the View/Modify option. In the reader setting select the COM port from drop down list and choose as you have seen in the device manager that was port COM9 (as per your system) and click on ok . I hope this may help you.
