Actually it is independent. You should use default Metahuman for beginning but still you can take your custom sculpt and work as static mesh input. At the end you will have the mesh you desired. So your questions easy and clear answer is; You can work with your custom sculpt.@@SushiMoog

Does this pipeline work on custom metahumans (ie the ones made by importing your own sculpt through metahuman creator) or only on the default ones?
You can use any shape for custom mesh but for the base skeleton to start with, you should use default Metahuman

can I use this to convert DAZ characters to Metahumans and keep their topology?
Currently it is little advanced but for sure it will be an easy process in further updates
