  • ( www.abnova.com ) - The thawing procedures for regular (non-DMSO-sensitive) cells are performed. The cell thawing procedure is stressful to frozen cells. Time is the most important factor. Thaw frozen cells rapidly (less than 1 minute) in a 37°C water bath and work quickly can ensure high survival rate of the cells. More videos at Abnova www.abnova.com

Is this applied to cancer cell line too?

Yes, you can use the same cell thawing procedure for cancer cell too!

How I kmow DMSO will be under 1% after adding cells? How to do the calculation?

To ensure DMSO is under 1% after adding cells to suspension media.
We are assuming that the freezing media is made up of 10% DMSO, 90% FBS.
      Identify volumes:
      • Let V1 be the volume of cell suspension.
      • Let V2 be the volume of cryovial freezing media added.
      Formula:
      • Final DMSO concentration = (0.1 x V2)/(V1 + V2)
      Inequality for DMSO < 1%:
      • 0.01 > (0.1 x V2)/(V1 + V2)
      Simplified condition:
      • V2 < (V1)/9
Add less than one-ninth the volume of the cell suspension in freezing media to keep DMSO under 1%. For example, if you have 9 mL of cell suspension, add less than 1 mL of cryovial freezing media.
