For example, if i prepare 5microgram/ml is it i need to do half-half means 2.5microgram/ml of isoniazid and 2.5microgram/ml of pyridoxine and put into a same cuvette then i run the sample to get the absorbance. Is this the correct way to do accuracy?

In the review discussion, comparing the truth of the obtained levels only or compared with other modern spectrophotometric methods or better with more sophisticated ones such as HPLC and to compare with HPLC the discussion only looks at method validation? But others aspects like cost effectiveness, ease of method, sample pretreatment, time of analysis, etc. should also been taken into consideration while comparing. The unit of concentration after solving the equation depends on what unit u have used for calculating absorptivity i.e. (ax1 for eg. let's say if a standard solution has given A=0.2435 and has conc. 10mcg/ml so absorptivity will be A/c = 0.0243) in this case the last conc. after solving an equation is mcg/ml. If for calculating absorptivity for the above solution and if we are converting this 10 mcg to mg (i.e. 0.01mg) then the last conc. unit will be mg/ml.
Sir is it possible to solve these simultaneous equation to find concentration by considering absorbances at any 2 random wavelengths without considering absorbances at each other's wavelength of maximum absorbance?
It's a very good question. Yes, it is possible, however, it doesn't assure u to get accurate, and linear concentration. On the other hand, if u record the absorbance at its lambda max instead of some other point on the absorption curve it gives accurate and linear absorbance on which u can relay. Additionally, absorbance recorded at lambda max will also reduce the interferences coming from the instruments which otherwise are more when recorded off the lambda max and may give the false concentration of ur analyte.

Yes you can use this method, 263 and 290 will the lambda1 and 2 for these drugs in water as solvent. Just check the dilution factor rest all is ok. Thanks for watching this video.

@@nursuhailabintisudarman9840 yes u can record spectrum of both the drugs (stds) seperately and then overlap the speactras and find out the Isobestic wavelength. Once u get isobestic wavelength then u can record the absorbance of ur formulation/ mix using photometric 8 lambda/ multi wavelength photometric mode of UV 1800 (one will be the lambda max of either isoniazid/ pyridoxin and other will be isobestic wavelength).
Sir, how about simultaneous estimation two component/multicomponent by area under curve method, It's the same with this? And spectrophotometry AUC method the analysis using cramer's rule and matrix method means?
The AUC method for multi-component is quite the same as that of the simultaneous estimation method, the only limitation of the AUC method over the SM equation is accuracy and reliability. Determination by using Cramer's rule and matrix method is the separate method using a mathematical equation.
