@@almche8978 yes u can use dmso for solubilizing ur sample. Just try to absorb on silica and then evaporate the dmso and load the sample in column as shown in this video instead of loading in the liquid form. Thanks for watching.

@ch.harshart1457 व्हॅक्यूम rotavaopour use करके concentrate कर सकते हैं. Liquid sample also can be loaded, not mandatory to convert to solid only.

@@brindhalakshmi2241 You can make the dilution in such a way that both the drugs should give absorbance less than 1. Usually 20000 to 30000 times dilutions are preferred from the original sample. But as in this case sitagliptin is 50 mg and metformin is 500 mg u can try doing 40000 times dilution and see. Best wishes Thanks for watching .

To avoid any false result due to solvent, it is used in control group

That mean in case of brain higher areas are affected first and then the lower areas like brainstem. But in case of spinal cord anesthesia spreads from lower end to higher segments of spinal cord. The last area of CNS that is affected is medulla which has vital centers. Hence anesthesia should be kept light or else vital functions will be lost.

It's very important to remove the moisture (which is adsorbed as impurity on the sample) so that it will not affect the separation/elution. Additionally when ur adorning sample on silica presence of moisture will not give u dry free flowing powder for sample loading in the column. Thanks for watching.
