- Welcome to our suite of NUI Galway Microbiology teaching videos. We hope that they will guide you in performing your experiments in a scientific and safe manner.
This video details how to perform a bacterial smear. This is a critical part of any bacterial staining technique.
The smear is heat fixed to ensure the bacterial cells stick to the microscope slide and to kill cells on the slide.
Steps:
1) Label glass smear in corner with culture name
2) Transfer small drop of water aseptically on center of slip using wire loop
3) Aseptically take a small quantity of a bacteria colony from agar plate
4) Transfer bacteria culture to water drop on slide, and spread around till the water doesn't look clear
5) Allow smear to air dry
6) Once completely dry, heat fix smear by passing slide (facing upward) over the Bunsen flame 3 times.
