Traditionally, yes. But since he is using PH the glue is more of an assist, and it will penetrate the finished face a bit through the screws entering. But if it were me, I'd have definitely scuffed them a great deal. But I'm also not a fan of screws. I use PH jigs, but instead of screws I prefer ⅜" dowels. Plus, if I'm not painting it (which is the only time I buy prefinished plywood) it looks beautiful if you can use a dark walnut dowel as your joint, and make it exposed. But it would be moot for this dresser build.
Im a contractor new to woodworking. I need to build a big 70 inch wide 40 foot tall closet built in and im lost trying to figure out where to start and how to make it easier. Im so screwed

I thought you had to scape a prefinished surface where the wood glue will be in order to get proper adhesion?

Why are you applying wood glue to finished wood?
Wood glue doesn't really stick to Pre finished plywood. you should really sand that off.
