    How many times can the biocoupler be autoclaved before it starts deteriorating and contam rates increase? I assume the frame is made of PP? How often do you have to replace the breathable membrane?

      Many BioCouplers have been in regular constant use for several years with no degradation with standard autoclaving, i.e. 121C (15 psi) for 20 minutes. The material is a proprietary high-temperature plastic that is rather hard. It resists breakage. If accidentally dropped to the lab tile floor they bounce, rather than breaking. The seal gaskets are silicone rubber and replaceable if ever needed. With care, the vent filters can be used multiple times but considering the value of the cultures it is wise to replace them after 3 - 4 months of repetitive use. They are inexpensive and easy to change. The BioTilt machine is made primarily of anodized aluminum, stainless steel fasteners. The "arms" have long-lasting neoprene-type rubber edging that grips and cushions the BioCoupler vessels. This system is economical but is of commercial lab-grade quality and built to last.

      The filter has to be replaced when it gets dirty. We usually get between 3-5 uses.We haven't seen any changes on BioCouplers with more than 25 cycles.

    How is the design ensured that the air filter does not get wet? What happens if you turn the biocoupler upside down?

      As long as you place the BioCouplers with the filter facing up. Then you guarantee that they will not get wet.

    Can you please explain that the ms media that you used in the video is in the powder form, but I couldn't find it in powder form on your Plant Cell Technology's website. On the website, MS Basal medium is available in liquid state.
    So, my question is, is there any difference or just same

      The liquid media is easy to use. You don't have to dissolve the powder in water. Based on your experiment, you can just directly add hormones and agar and sterilize the media. So, the liquid media eliminates one step.

      @@ShaikhMuhammadNoor, a one-liter solution contains 4.54 grams of MS, and our bottle already contains that. So you don't need to do anything.

      @@ShaikhMuhammadNoor Yes, that's correct. You can add other substances based on your experiment. There's nothing compulsory. You can also use the media directly without anything. Yeah, please sterilize the prepared before using. We have many videos on media preparation on our channel to learn how this works.
