They can if your not in front of the door and it's not only on expert tanks can break safe room door on any difficulty
Tanks can break the safe room doors in the beginning of a campaign level if you retreat back into the saferoom and close the door
Once a saferoom door has already opened, It becomes destructible.

You press Mouse 1. And it has to be with a crowbar, sword, fire axe, or cricket bat. You also have to have your crosshair right on the tongue.
