Ehh that fan theory seems pretty wack imo. Mayuri is supposed to be an orochimaru like character who is balls to the wall sadistic and evil just because. It’s supposed to show how obsession drives you mad. I don’t think he’s supposed to have a sad/tragic backstory. I think he’s just a deranged scientist; it was a very popular trope back in the day.

Unfortunately anything that would truly explore and substantiate the theory would require very spoilery discussions about zanpakuto and their nature that I couldn't in good conscience do here.

He's actually based on the Japanese scientist from World War 2 that conducted many fucked up experiments on many live prisoners of war, not caring if they died in the process of his experiments and examinations. Kubo, like many other authors, used a lot of real world people as the inspiration for some of his characters.
Censorship in this particular episode (Source: Bleach Wiki): «Mayuri after being shot by one of Uryū's Quincy: Letzt Stil arrows was drastically toned down in the anime. In the manga, Mayuri was engulfed in flames and was missing his left arm and shoulder, his left pupil, and a portion of his left side. In the anime however, he was only missing most of his left arm.» «Mayuri after being grievously wounded after Uryū destroyed Konjiki Ashisogi Jizō was drastically toned down in the anime. In the manga, bone remnants were shown in the hole in Mayuri's body, whereas in the anime, the remnants are not visible.» «When Mayuri performs Nikushibuki and starts to liquefy, his eyeballs aren't shown in the anime as they are in the manga.» «The small, slatted portcullises with multiple earrings in place of where Mayuri's ears would be aren't visible in the anime as they are in the manga. Instead, they are obscured by strands of Mayuri's hair. Also in the anime, the scars that are on his lower body are not shown, leaving the ones on his upper body visible.»
My personal pet hypothesis, is that any creation has to start with a "base" soul. It contains everything a normal human being contains... emotions and such. Mayuri strips a soul of the "dis-likable" properties and then puts it in a body that is made by growing his own cells. He is just not a likable character... though he is a memorable and interesting one.
If you get to the point I will be referring to remember that little note you made about him making something imperfect. It's nothing major and it's far down the line, but it's fun that you used the term imperfect.

Letzt Stil gives Quincy an immense boost in power at the expense of them losing their powers.
