    @Microbehunter : In your comments, you mentioned that "wet mount, or permanent mount, uses water as a mounting medium". A permanent mount doesn't use water specifically as a mounting medium - it uses a mountant such as Canada balsam or another resin (including Numont, L&C, etc), although water may be a solvent for such a mountant (e.g. L&C).

    Been wanting to get into wild culture ranching with acetobacteria and yeast, and I've been liking your channel. As a child, I used a miniature microscope to look at macroscopic objects, but the more methodical, proven methods you're using on your channels are very helpful.

    I love your simple and effective tutorials. Might I add another method for observing large specimens? Get a ball of soft clay, take your cover slip and gently scrape each corner across the clay to accumulate a tiny bit of clay which will act as little feet and raise the slip when placed on the slide. This was how it was done by my biology 101 lab professor. Cheers!

      Another strategy is to put some tape on the sides of the slide and mount the cover slip on it

    Sometimes I do take some water off, but can't help thinking about all the protozoos that might be going off with the water

    Are single depression concave slides used for wet-mounting too?

      Yes, especially for water fleas and other orgamisms that are larger and which might be squashed by the cover glass. For smaller organisms concave slides are less suitable, because the water organisms swim vertically in and out of focus and it is difficult to track them.

    can you make a dry mount with an originally wet specimen?

      Problem here is that the speicmen might shrink and deform when dry.

    What microscope can you take right up to the water's edge? (no samples)

      Generally any microscope that is a. sufficiently portable and light and b. has battery-powered LED. You need a so-called field microscope and there are many varieties of them. Google for "field microscope".

    my cover glass has been floating because thy soil pices are too big sometimes

      You can stabilize it by making a "bridge". 2 cover glasses (left and right with space in between) hold up the third one.
