The Dirtiest Things In Your House (You'll Be Surprised!) | Bacteria Experiment

  • Here is a link to the agar kit that I used so that you can do yourself! Last I checked it is only 18 dollars.
    www.amazon.com/gp/product/B00...
    In this video I show you just how dirty common household items are. There are bacteria everywhere! There are about a million bacteria cells per milliliter of water. There is more biomass of bacteria on earth than the combination of all plants and animals. In the video I test common household items to see which items harbor the most bacteria. In my experiment I spread my bacteria samples on tryptic soy agar plates. I sample the kitchen sink, the toilet, iphone, floor, doorknob, remote, mouth, toothbrush, my son’s stuffed animal puppy, and my skin (my armpit and belly button). Then I let them grow for 12 hours, and count the number of CFUs (colony forming units) there are on each plate. I do the same at 24, then 48, then 72 hours. After each time increment I count the number of bacteria colonies. The end results are shown below:
    12 hrs 24 hrs 48 hrs 72 hrs
    Sink 3 10000 10000 10000
    Toilet 1 39 372 600
    iphone 0 1 41 74
    Floor 0 0 63 97
    Doorknob 0 3 11 24
    Remote 0 1 1 42
    Mouth 0 0 950 1500
    Toothbrush 0 0 0 1
    Puppy 0 3 96 222
    Skin 0 0 1 8
    In the end the sink is by far the dirtiest place that was measured. The whole area that was swabbed on the agar plates was completely covered by 24 hours. The next was the toilet. The surprising results were that my skin and my toothbrush were the cleanest out of everything with my toothbrush only having one single colony of bacteria on it in the end.

    I was always taught in microbiology to keep the lid over the Petri dish at a 45 degree angle so you won’t contaminate it with the germs from the air or mold.

    • @veratisium
      @veratisium 2 года назад +6

      @@xmipad-rafaelxaverianmulya6084 what he means is you dont open the lid completly if you want to put some bactirial spore or fungi spores. So when you want to innoculate something you open the petri dish but the lid in your hand covering the dish as much as you can and you innoculate the dish that way, in order to not get some unwanted spores from your envoirment. It would be to innoculate under a Still Air Box or Laminar Flow Hood but thats quite expensive. But as always be as clean as possible and the room or envoirment clean as well and "settled air". No AC or windows open because the air will make the spores flow in your room. Sorry for some broken grammar.

      I saw where you can carry the objects, when possible, to a sterile table area with a bunsin burner lit to just barely lift the lid off when you swab your q-tip sample to prevent the Air from your mouth & the room from contaminating the sample.

    I always spray clorox foaming bleach cleaner (or it's equiv) on the the surfaces of the sink, counter tops, toilets, showers, bathroom counter tops, etc and let sit for >10 min to obliterate most of the bacteria/fungus/molds, then scrub and rinse and once I am finished cleaning/disinfecting, I spray everything down with Lysol (or it's equiv). I wipe door knobs, phones, fridge door handles, remotes with clorox wipes (or it's equiv). I always mist my toothbrush and hairbrush with 91% alcohol and let them air dry. After I use the bathroom sink- I also mist the sink and faucets with 91% alcohol. It's exhausting to be an OCD inflicted clean freak- this all started of course with my diagnosis of OCD, but what triggered my cleaning mania is taking a college microbiology course. It ruined me! LOL! PS I know I need help :)

    I think that there is a lot of bacteria in your mouth, but the specific species there don’t grow well on Tryptic Soy Agar, and some of them are fastidious, in that they require specific nutrients you would find in your oral cavity. A lot of streptococcal species behave this way, to say the least.

    I worked in a major pharmaceutical company in allergenic extracts. In the clean room with hepa filtered air, positive air flow, and wiping the filling room top to bottom in isopropyl twice each shift they sometimes STILL found bacteria. The QC
    people would swab door handles, doors, windows, and equipment touched by personnel. They also would swab places out of reach like the ceiling, above the door jamb, STILL they found bacteria maybe once a week. If a clean room has bacteria, everything in your home has it. Now do a video on dust mites and other evil things lurking in your home. Its worse than you think. And the creatures living off your skin. Our crew watched a video on bacteria an a co-worker threw up. I almost did.

    This is an interesting test but you did not use sterilized water ( and it should also be saline water to not cause a shock to the bacteria), I also think the way you spread the bacteria on the agar plate is not representative of the surface.
    But it is a fun experiment just to get a little idea of what is growing in your house :)

    • @jennyneedsmeds
      @jennyneedsmeds 6 лет назад

      I believed that you're supposed to keep the sodium levels as low as possible in this type of experiment... as salt hinders bacterial growth. Which is true? I want to try this, but I want to do it right

    • @finalbossd
      @finalbossd 5 лет назад +3

      j money Microbiology Student here.
      It depends on the type of bacteria you are looking for. Some bacterial genera like staphylococcus like having high salt levels, so to isolate them, we use a type of agar called Mannitol Salt Agar instead of the Tryptic Soy Agar you see in the video. In fact, when you pickle things, all you are really doing is just creating a favorable environment for certain bacteria that you want to ferment your food while making an environment unfavorable for bacteria that spoil it.
      In general, you may want to use a low saline solution, since using distilled water can be too hypotonic for the bacteria, but you would want some amount of salt.

    So I am writing here, because I thaught you maybe see my message here. Here you do not have so many followers and views on your vids so maybe you recognise me.
    I have a Idea for your Vacuum chamber:
    Laserlight produces heat when it collides with solid molecules i guess. So if you glow with a laser in air you maybe seeing a warm line through a heat-detecting-camera (wärmebildkamera in german). This is because of particles and molecules in the air.
    So what if you did this in the vacuum chamber? There are as less particles as possible which would produce heat when they get shined on. But there are none. So would you see the warm line through the heat-detecting-camera(german: wärmebildkamera)?
    I would like to test this out but i have not the supplies to do it;) thank you and much fun
