Big plant been real quiet since this dropped.
😅😅😂😂❤
This will be extremely useful for growing cactus from seed. When we grow cactus from seed, we typically grow it under cover for weeks or months or even years, so the seed doesn’t dry out and die or get stunted. Cactus seedlings have minuscule root systems and it takes ages for them to develop. Normally we sterilize the soil in the microwave and then grow it in some kind of plastic container with a lid. I’ve had a lot of failures with this method due to mold and algae finding their way inside and either suffocating the seedling or causing damping off. Next winter I wanna abandon my caveman ways and do it the proper scientific way.
Since cactus seeds are soooo tiny, you could also use a micropipette with sterile tips to pick up the seeds in the solution and place them directly onto the media. I am planning to try that soon with some gibbaeum seeds
I just started growing cacti & various succulents from seed - great idea!
She makes me want to go out and get the gear and try this!!
Thanks for bringing joy to this retired plant geek!
thank you!
I guess I'm a caveman.
Fr
I'm apparently part of your tribe then. Lol
Guys, forget them. Let's go walk in the manure.
*Picks up Spear and Leaves*
Ouga ouga !
Whole Neanderthal right now
Ah the pressure cooker. The smaller humble cousin of the autoclave. Or you could call it the home autoclave I guess!
Back when I worked with mammalian cells we would give them a bit of penicillin/streptomycin to reduce the risk of contamination. It's probably poor practice due to the whole antimicrobial resistance issue we've got going on, but it allows the students to not contaminate all of their samples.
OMG OMG OMG
An affordable flow hood!!!!
ty ty ty 🎉
This is my favorite channel
Love her sense of humor and enthusiasm.
This is next level gardening. yikes
Can you germinate cactus seeds like this?
Yes 100%. But since the seeds are so small I would use a sterile micropipette to transfer them instead of forceps.
At the rate this is progressing, we'll soon get a video showing how to extract and culture your own bone marrow stem cells using kitchen utensils and a pressure cooker.
Don't give her ideas. Please! 😉🤣
Thank you for this video. I have no interest in all the cloning and stuff, I just like your channel and the processes you employ (I'm a nerd). The seed starting though! That's new to me and exciting. It's a different way for me to experiment starting seeds for my garden. I begin the process every January, so I have time to prepare. Thank you ^_^
Your teeth look like diamonds wtf
This is one of my favorite subscriptions;)
Great video and sounds perhaps the best way to get into tissue culture before going the whole hog of trying to sterilise explants etc.
Any plans to do a demonstration video with the mini flow hood? 👀
Probably in my next vid! I wanted to use it for this one but I filmed most of the video before I got it
It just dawned on me that your channel would literally blow up after a nuclear war..........................................................................................................................
I've always used polypropylene plastic bags for sterilizing tweezers and other hardware. And you can re-use them for 10's or even 100's of times.
you used the cotyledons not the leaves
When I first learned how to do this I had the biggest nerdiest smile on my face thinking about how I could use this method. I don’t have the equipment since it was the companies equipment and I wasn’t really looking because of how expensive it typically is for their methods. Though seeing your video makes me want to get back into it and buy the stuff for my own garden.
A bit of a germaphobe aye
Dang that baby laminar flow hood is tempting!
Excellent 😃🎉
I subscribed to your channel 🙏🌱
Thanks for another excellent TC video!
Anyway, a question. How long roughly from initial seeds to planting seedlings in the soil? This would be the seed - explant - cultures - soil option and for sunflowers.
Great video 🙏
Appreciate you!
Thanks for the video! Can you give me a hint for my containers pls? I sterialize them in the microwave but as soon as it cools down they suck the lid and the container gets dents from that
Great video.. thanks from the UK.
This would be great for difficult to germinate seeds.
I would have loved to see the cutting procedure.
I was expecting you to use the baby growth point, not the leaves.
I came here to learn about plant tissue culture techniques, and stayed not only for the marvellous content, but because you are the sexiest, funniest nerd I have ever seen!
I used to love watching the Big Bang Theory, and I have to say, if you took the three leading ladies ie Penny, Bernadette and Amy aka Melissa Rauch, Kaley Cuoco & Mayim Bailik and somehow amalgamated them together, you would get YOU!
Beauty, pazazz and brains.
I know these days that you aren’t supposed to say things about peoples looks etc, but I just had to! Also, your humour is off the charts dry and funny as heck.
Thank you for posting all you do.
Content, 10 out of 10.
You…., You destroyed the scale!
Your videos are a treasure! One year ago I watched your DIY autoclave video and next week I’ll be starting a job in plant genomics. On paper, I’m completely unqualified. But the knowledge I’ve amassed from your channel gave me a shot. You’re making a huge difference if a shmuck like me can get into these spaces. Keep up the phenomenal work!
When germinating the seeds, what media did you use?
Or use indeterminate varieties then take cuttings 🤷♀
Great video! Also, where is the link for plant cell technology?
@@IronAlien Just updated the description to include all the links :)
@@plantsinjars Thanks 😊
Amazing video! Do you ever wanna do cacti?
Great video 👍
I was surprised the cotyledon leaves were used vs the first true leaves. looking forward to the longer x100 vid , thanks.
Would RO water work?
I love this resource. You're a G
best intro
coolest vid ever
I really enjoy your channel. I don't think that I will ever try to perform any of the operations that you do on tissue culture or plants, but it is fascinating to see how technology can improve productivity.
amazing video!! one question, 5ml of h2o2 and 45ml of water gives 0,3% of final solution, that is not very weak to sterilize? usualy I like to use 1% solution
I'm doing classic gardening and it does feel caveman like in comparison to TC. What would you say would be a good application for the technique you ahowed here? I'm thinking about expensive seeds, but I'm not sure if TC would come out ahead for garden plants.
Very interesting technology. Ever since I got to know Kallus cultures, I have been fascinated by the possibilities.
It's also more fun than with animal cells, both ethically and technically easier!
This is alot for such cheap and common plants have you tried growing mushrooms you have the right set up for mycology
Interesting to know how easy this process is once you have all the equipment and knowledge. Thanks for making this video to educate us.
However, I don’t mind waiting a year to collect seeds from the best plants as that’s what I do every year anyway.
Any seed?
I would think this would work for any seed, with some modifications. Note that some seeds require stratification to germinate - you would cold treat the seed and then follow the procedure.
Do you find those polypropylene pots crumple in the autoclave ? The company selling them advises against sterilising with the media in them
What are you studying faculties, so you start up making this beautiful masterpiece stuff?
I really love your humor :) I am inspired! I'm gonna try and grow some Wasabi seeds using TC
I bought a Hanna instruments pH meter it lasted exactly one month past the warranty and they didn't offer repair service.
Whose pH meter lasts?
hahahahahaha that intro xD
Dr pepper tofu flavor !! Omg !!
Nah bruh lavender honeysuckle sprite
Now i need to taste this !! Only in japan
When you do the replicating step. How long can a plant stay in there before you are forced to do something with it?? Just wondering how often work needs to be done while rotating a catalog of plants over long period of time.
@plantsinjars can you do a video about engineering glow in the dark house plants?
Great channel, fellow botanical nerd here. U are far more advanced in Tc than me. Lol I started here and ended up in fungal tissue culture but still love plant tc
I'm curious about trying gibberellic acid to germinate old seeds.
I loved the video, I'm going to try to do it at home
This is amazing.
I can see the applicability of this technology really only to rare or exotic seeds.
Aluminum foil is the street method?? We used that all the time on class!!
U Should ad to the title 'part 1' so you end up with the 100 on the last part
Have a great Month!
I find autoclaving typically crushes polypropylene containers, do you find that not sealing them helps?
The larger ones sometimes do get crushed if you stack them. If you have a laminar flow hood, you can pour the media under the hood. The small ones I use from lollicup also do not crush in the autoclave, even when stacked. Hopefully that helps!
Man I need to find someone like you wow love your energy
Wanna see waterlily tissue culture from tuber
I am addicted to isopods I breed them lol
-actually a cool biology/engineering/technology video that might be useful
-linked products
-in vid ad, followed by yt ad
-good thumbnail
-literally can sell the results of the video (ok maybe sunflowers are whatever)
Bro is grinding
broster's about to ride the algorithm to 1m subs
JUST FOUND YOUR CHANNEL AND I AM GLAD
It's all about the vibes. In a mission to prove my mechanostimulation technology can bolster plant steering processes.
Great vid... Cheers!
Soooo. I'm gonna stick to collecting seeds and taking cuttings. Still get 100's of plants from one tomato seed just by letting it fruit and produce seeds. Or whatever plant you're growing. This seems.......
Have fun I guess! 💜 happy growing 💜
Im trying to be better Laur!
Hello, I would like to know, where did you get these nice culture grow racks?
The shelves? They are from Home Depot. Amazon also sells similar ones
@@plantsinjarsohh really, whats the name? i was searching for shelves with lights but i didnt find any good ones
@@mochartsmo2122 I bought 4 ft lights from Hortipower and attached them to the Home Depot shelves. Hortipower sells grow racks but they are extremely $$$
@@plantsinjarsDo you use the Hortipower Nuser 1, 2, 3, or 4? Also, would you suggest for growing microgreens?
Nurser 3. I don’t grow microgreens, but I don’t see why they wouldn’t work
Yaaaaaasssssss 🎉🎉🎉
Hi could you tell me can banana peel be used as tissue culture if yes how?
Probably not but I have a banana specific video in my writing drafts
“The street method” lol
Skoog as in fugue
Very dumb question, how much water is in the pressure cooker when you sterilize things?
Depends on the PC, but mine is 12 cups
@@plantsinjars Is there a rule of thumb with the depth? Or is it something that would be specified in the manual? Also thanks!
Smart girl!
I was wondering if there was a reason you chose to use solid media instead of the martini shaker method?
Your cute❤
❤️👍👍
❤
😍🥰
Does plant cell technology have issue with purchasing supplies often. They have been out of MS forever?
It will be back in stock so soon!
I have to say this is the video that woke me up to this❤. I'd seen so many tissue culture videos on FUNGI, not with plants, at least by how I was searching. My first experience with it was really at Disney World, the place known as pleasure island, had some really cool stores and they sold sequoia trees in testtubes. and of course they were adorably tiny in there and I wanted to buy one, but I was afraid I'd kill it as I didn't know sooo much at 15 years. I think you're channel is a gem and your vocal delivery has a jovial personality that brings an inner child to life within me as I watch and learn from you. May you in all facets of your life know true, blessed joy.❤❤
which disney world did you find that tell me I HAV 2 GO!
@Micheal-uc5qb I used to live in Orlando, Fl. If the Virgin Music Mega store is still there it's in THAT shopping spot (Pleasure Island, adults only when night falls. This was 29 years ago. I'm old now!)
I’ve had this thought in my head a few days… if you add two types of the same plant like say Thai constellation and a albo monstera could you cause a hybrid in tc? Maybe if the tissues are “grafted” to each other might help get some cells to swap. Just a thought, could be a cool experiment.
That’s exactly how it works, it’s actually a breeding technique that already exists. What u described creates a chimeric cross. Although not a hybrid because the plants u described are in the same species. U could take it a step further by a another method known as “Cellular Fusion” by coating the cells with polyethylene glycol, which dissolves away the membrane, letting the cells merge, replicating natural hybridization. Nowadays u don’t need to wait till maturity.
Naturally cells will not merge unless the membrane is penetrated… if u remember in school… reproductive cells has the ability to penetrate the other team thus allowing fusion, while with vegetative cells that layer has to be removed using various ways.
@@Micheal-uc5qb that’s really cool thanks for the info, I want to try it now!
@@Micheal-uc5qb now I’m dreaming of a pink variegated monstera. Thai x pink princess maybe 🤔 would that be possible?
@@mil0thebird possible, u can even do it without TC. Find a Thai constellation monstera stem with a bud cut off the stem just right in the spot it's growing a bud clean and straight with precise tools, try your best to make an exact half cut of the bud-junction and do the same to the pink princess. Connect the top half with the bottom half. And apply keiki paste on the bud eye. Now take good care. pruning off other potential growth points may lessen the wait although not nessasary.
Hey there, love your work. I noticed the Dr pepper, sugar free. Please drink regular as sugar is way less toxic then aspartame. Thank you. This only applies if doctor pepper is using aspartame. If not disregard this message. Have a great day!.
Is it safe to heat up the plastic cups
Yes.
Maybe use glass instead.
There's plenty of autoclavable plastic.
