In the last battle, your opponent was a cold build (the two staves). This means he slows down your backpack, including the weapons in your backpack. Because of this, your weapons use your stamina slower, giving time for your stamina to regenerate, and that means you didn't get to use the potions you wanted to use. Then you reached half health, that's the condition for his potion to trigger and damage you based on your debuffs. You had 75 cold, you took tons of damage, and lost the fight.
Useful tip: You can rotate items, including potions. If you rotated your potions so that they were horizontally in a column, you can get a good long chain of dual effects going on. Very effective when you mix in healing ones. You can also get a potion to lie in two potion belts to get the extra benefit twice.
You got to merging gems consistently and that was great, but the Torch was the same mechanic. If you hover over and item that can Merge, it will show a disjointed blue line to the other item it's synergizing with. Even if it's in the shop, it will show if it's possible to combine.
Its fun to watch but I think what hold him Tyler back was that he ignored so many merge or interactions. For example, it seems like the banana and potions had something going on. He played it like Backpack Hero, where organisation is key, while here it seems interactions are more powerful
Useful tips: if you hold "ALT" on an item it will show you the recipes of it, after each round you can click on "hide" and then on "logs" to see all the stats, if you mess up your backpack you can "ctrl+z", if you click on the settings wheel you can see a few controls and if you click on "return to title" you can close the game and continue it later from where you left off, if you click on the "items" icon you can see them all together with their recipes, and you can test your builds in "History" if you want. Game becomes really fun when you start getting the hang of it, consider giving it another chance.
by the way d.o.t damage is busted in this game, and yes the panel don't show stamina regened by potion and food item. plus the shield has a cool mecanique with some good special effect that let you negate part of the damage
This game has been my go to for a while now, really nice to see him play it! I just love how each run you end up with new synergies and a new build to try, even after like 30 runs or more. Oh, and if you hide the victory screen, the combat log is really extensive and even has graphs

Idk if you've seen the game but there's a really old resource management auto battler called jack and Cassie. It's on gamejolt I think. It's only a demo and for certain won't ever get a full release, unfortunate situation with the dev. For what there is, it's really good.
Tyler should definitely look out for the upcoming game "The Bazaar". It's similar to backpack hero and this with asynchronus multiplayer mixed with PvE battles and a roguelike map. Tons of synergies and I'm sure Tyler would cook.
Having played a lot of games like this, I'd like to suggest Backpack Brawl. It's very similar to this, but: + Is free, has more items, there's twice as many characters. = A leveling progression system that encourages you to play more to earn level ups for your characters and unlock new items. - Microtransactions to unlock characters (Though I earned enough in game currency for free to buy all of them within about a week of play.)
There is a game that is shockingly simular on mobile, being Backpack Brawl. I would assume that Backpack Brawl was a knockoff of this game, since it is more advanced. However, it is very unlikely for a knockoff to have more polish than the original game. Also Backpack Brawl came out first I think. And it has a build where you can fill your entire backpack with nothing but rats.

I think it was also part Fatigue build, Based off of the hundred and seventy five healing, they got when night set in

Tyler it is generally more efficient to put the potions sideways in two potion bags it increases their chances of activating multiple times
