Music theory nerd here! I'd say this riff involves a modal mixture between D major and D minor. The riff uses two chords, D major (I), and C Major (bVII). C Major is not found in the D major scale, but it is found in D minor. The chord is also found in D Mixolydian, but Iron Maiden mess around with an F note which isn't found in the scale. Quite simply, they basically took the notes and moved them down 2 frets. So while calling it D major is partly true, there's more to it than that.

The whole song uses both D major (D Mixolydian actually: the vocals use C natural and F#) and D minor actually, although in other sections the switch is more evident.

No, not the song as a whole. It's a D#/Eb drone. It's in no "key" really. It is meant to be chromatic and/or atonal. The only real constant is the D# as the tonic and the A# as the fifth.
