    How do manage to not have an air bubble in the Durham tube when you are setting up the experiment? I cannot understand how you can ensure the small tube is filled with the sugar solution and manage to get it upside down into the test tube without leaving an air gap.
    I would be interested in your clarification as I would like to set this experiment up for identification purposes relevant to beer microbiology. Any reply much appreciated. What am I missing?

      Add the small tubes to the large tubes, add the broth, and autoclave. After autoclaving, the gas tubes will be filled with broth.
