Finally we call that Net specificity of screening covid 19 negative passengers was high in the particular flight but based on sequential testing we identified quite few COVID patients.

In case of net specificity why wo didn't add, unique patients identified by each test as true negative, with the common true negative patients ,just like we did in net sensitivity case, while calculating net specificity? Instead we just took common true negatives of both tests🤔
this depends upon whether you are opting 1) Sequential OR 2) Simultaneously screening tests as in sequential testing objective is to exonerate maximum negative while in simultaneous screening we want maximum inclusion.

What if you perform the same test twice? Would the specificity and sensitivity for the second time be the same?
It may vary too, as depending upon sample condition (degraded RNA/ DNA etc). Hence we repeat assay thrice to refine our understanding (range values)
