My favorite theory is that gray place below the game is the game engine and the Wreckingball and Meatball Man are assets waiting to be instantiated when their level comes around. Runmo using Jumpmo's head as a 1-up caused a glitch in the game, releasing Grobbletombus, a removed feature to wreak havoc on the game.

There was a neat line said by the glowy thing that went "The pursuit of life is more dangerous than a life in pursuit", which could probably relate to some... Kinda message, probably? You could also say the message is to not to drugs because that's how I felt watching this

It relates to the main message of the story. Runmo feared death to the point of abandoning the adventure set for him in life; his "life in pursuit". He desperately tries to find more lives to secure more time for himself (the pursuit of life). In the end, he gets more lives and tries to safely continue his adventure, but it is too late and he is out of time. His pursuit of life cost him his adventure; his own chance to find fulfillment. It is a tragedy where Runmo is so scared of death that he misses out on life, and serves as a warning for us not to do the same. Or not idk that's just my interpretation.
