    Dear Mam, you used autoclave and flame that's good but, you didn't used Laminar air flow. So there is a risk of contamination.

      Agreed. It should've been done in fumehood.

      But autoclave has killed the bacteria on the surface of glass tubes.. That why we didnt used laminar air flow..

      @@IScreenshotNFTs Fumehoods are different from laminar flow hoods, I just learned about this recently myself. Fumehoods are meant to pull air from the room, into the hood and are exhausted out to reduce chemical fumes for the person working. LFH pump hepa filtered air into the hood and out toward the operator to reduce microbial contamination from the room air

      @@justinclark5844 Regardless, its done near a flame anyway. Doing in Biosafety cabinets is rare, because labs also do mammalian cell culture

    why before closing the bottle where the culture medium is passed through the flame of the fire?

      this is for sterlization, to remove any present microorganisms or reduce risks of contamination.

      Dear, we used shaker for proper growth of the bacteria. Because when media shake then bacteria easily get their nutrient content.
