We have worked with S.aureus very often in our practice lab without any real precautions.
I do the same thing and still doesn’t work!!😢but now i know my problem maybe i didnt let the loop cool down

In what quadrant do the isolated colonies appear in the streak plate:
fourth quadrant
1st quadrant colonies are confluent, 2nd quadrant colonies are less dense, 3rd less denser, 4th less denserer
It actually Depends how heavy the primary Inoculum is. You should have isolated colonies by the time you get to the 4th quarter of the plate
