      If the determinant of the controllability matrix or observability matrix is zero then the system is not state controllable and not state observable respectively

      Here we are not interested in finding rank of the matrix. Our interest is to check whether rank of matrix is equal to number of rows in the matrix or not? This can be checked by finding its determinant as done in the video. To find the rank, you need to convert the matrix to normal form and then get the rank.
