In southern blotting we find a gene of interest from target dna for which addition of probe is required n then additional process like autoradiography ( incase of radio active probe) where how will you use this gel? If you are familiarize with how initially we loaded the dna you would understand it's not practical and i don't think it's possible for probe to run over gel again.

Leonardo Buback Yes, a 1kb difference in DNA fragments would be easily resolved by gel electrophoresis. I think a 1% gel (1g agarose powder per 100 mL 1X TAE), ran at ~100 volts for 30-45 minutes would work beautifully. You would run a ladder to gauge the sizes of bands based on their migration. I would use the 1kb ladder from New England Biolabs.

How is the nitrocellulose membrane helping in the hybridization? Why is it used? Can't it be done directly on the gel?
