Regardless, he doesn't lose interest in her after her reveal as a female and, for what it is worth, is enthusiastic about such a prospect. Now that doesn’t mean there aren’t other scenes where Yosuke is written badly, displaying homophobia and/or being mean to Kanji (looking at you camping scene) but this is not one of those scenes.
Yosuke is an accurate representation of a high schooler born in the early 90s. Virtually nobody from that time was acceptably woke by today's standards at 16, he was even well above average in that regard.
