    (review follows) here's a very nice video of a human hand doing a series of things with a precision pipetter and some small titration tubes. From the distance (and camera lens used), it is completely impossible to garner any sense of what is happening, other than to read the text. This generally makes this a text reading exercise with nice background music.
    for any given moment, is something going in or coming out ? here's where we change pipetter tips to avoid cross contamination, here's where we put in the "ingredient x"... try replacing all the captions with the words: here's me trying out my new pipetter i got for xmas !!! and see if the video is one tiny bit more or less informative.

      I completely agree. Trying to research RT-PCR and I'm finding it difficult. Thought I'd try watching videos but without captions, I have no idea what's going on.

    why is rnase used seperately if depc water is already in use?
    if we use normal water we can already get the rnase,

    quick question: why does RNA initially need be denatured? It's already single stranded- ready for the reverse transcription process.

      Pretty sure it's cos the RNA has probably formed secondary structures (bound to itself) so the high temperature will break the hydrogen bonds it's made between its own bases

      The purpose is to reveal the genes that express the mrna being reverse transcribed.
      And I know, four years ago...lol

      I know I am a year late, but if you know the RNA sequence, can't you figure out the DNA seq? or do are we making (figuring out the RNA sequence) the RNA using primers?
