I thought the explant will be excised from the apical dome of the meristematic part of shoot, plant in basic MS media with different supplements of growth hormones to induce it to form undifferentiated mass of cells or callus. I just didn't see this callus production and plant regeneration from callus in your video. BTW, do you conduct tissue culture training/classes and/or provide tissue culture services to interested clients ? Thank you very much
Could you possibly make a video on an Echeveria perhaps. Want to get into tissue culture for succulents etc... Can't find any videos. Yours are very helpful and easy to understand as well. Could you do one with leaves and main stem of plant? Or if you have information on" how to".. Found pdf's just want a professionals advice. Thanks in advance
When working with the apical meristem, I have got better results getting the plant established and then move to multiplication media. I think the Monstera was fully established after day 50. For leaves, you can go directly to multiplication media.
@@PlantCellTechnology Thank you for this thorough explanation. But with Monstera plant you cant start the micropropagation process only with leaves right? Is it only the apical meristem that works for this plant? Or would any younger node piece also work for this plant? With better results, do you mean a higher yield or lower failure rate or something else?
Hello, I started doing tissue culture on monstera too. But just wondering at what point do we know when the explant is ready for multiplication? Thanks!
Hi, thanks for making this video! I cant wait to see the multiplication video! Anyway, I am new and have some question here. I realised the media you used here is MS. Can I understand for multiplication purpose, we just need to add "rooting hormones" into the MS mixture?
@@PlantCellTechnology thanks for your reply! Will you be sharing the mixing ratio for multiplication and rooting? Hope you could make a video of mixing these 2 mixture too. Thanks.
I love this kind of video! step by step! Thank you so much! Greetings.
Great to hear!

Yes, we can add it to the list!
Awesome thanks so much
Could you go straight to multiplication media instead? Or does it need to be in the base media for a certain time? How long?

If is growing new leaves, you can move it to multiplication media.
@Plant Cell Technology Thanks! 😊
@@PlantCellTechnology Which is composition the multiplication media?
What is the next step, Once the monstera got the new leave and roots?
Next step is multiplication media!
@@PlantCellTechnology will you make a video about it?

For multiplication, we need a higher ratio of cytokinin compared to auxin. Then for rooting, we do the opposite (More auxin than cytokinin).

@@monsterahut2185 Yes, the video should come out in a few weeks.
how do you prepare ppm at 4%?
9.6 ml of distilled water and 0.4 ml of PPM + 4X MS salts. 4X MS salts for 10 ml is 0.45g
