Is there a way to know how much foam should be left to ensure the items stay separated upon moving or impact? I feel like it should be a lot, but that may not be the case. So for example, say I have 2 cylinders like a soup can but each weighs about 10 lbs, how much foam should be between them roughly?
The yellow part is basically a warning area. You can place you cutouts there. It means you are getting close to the edge of the case and as all cases taper inwards, if your shape is deep enough there might be a hole in the foam. Always check the 3D rendering to see if there are any holes.
The best knife for cutting PE foam is a fishing fillet knife. If not, any long blade that does not flex too much. Best way to glue is with a hot melt glue gun that you can get at any craft store.

What do you recommend the minimum spacing would be for example that harmonica case in between each harmonica.
I would recommend at least 1/2" but 3/4" would be better
