He's taking a concert Eb and bending it up slightly -- almost to an E natural, but not quite. After that he's hitting the Eb again and the next note is the C below that. It's a common blues device (playing both the major and minor 3rd), except that many players will take the bent note up fully to reach E natural. Because Lester doesn't (on purpose, I'm sure) he creates some momentary dissonance.

I open my grandmas phone and listen to one of the most beautiful tunes to ever be made. This inspires me to start playing the sax.

Never heard that quote. Great. I think the note was a little glissando on the minor third note of the "one" chord.
Blues is by definition a microtonal style. He is bending to reach that in between note.

Lester is my favorite jazz soloist, melody, quiet fire and the blues is a winning combination to my ears.
