That's cool 😊 thanks for sharing again! I just started my mushroom cultivating adventure last November. I got a lions mane liquid culture from you at the same time I ordered two of the 7lb lions mane fruiting block kits from MycTyson to make sure I could grow them🙂 They were fabulous to me, I got about two pounds from them on their first flush and just finished them off this weekend with the fourth flush about 1/3 of a pound just in time to put out my 8.5 pound block that's in my fruiting chamber from your strain yesterday, It's popcorned on the top and mycelium is growing everywhere else. I took a clone of the MycTyson lions mane to agar and it's ready to put to liquid culture now. I just want to say thanks, it's guys like you that I have learned soooo much, and I have done it all in a large still air box.

I use this method for getting spores from mushroom caps too... I got a tall container that I suspend a bread tie from the top with a magnet... Stick the bread tie through the cap and suspend it over the petri dish.... I get a lot of single spores to isolate for my monokaryon breeding purposes.

What about for home mycologists who don't use agar? Could you do that set-up, maybe with a smaller grill and a tupperware container inside a SAB--and have it fall onto aluminum foil? Would you then be able to make a spore syringe? You wouldn't get the different cultures to compare but maybe you could avoid contaminants.

But damn I just wanted to grow some lions mane for the first time, and sustainably. I want to do it properly, but I already have way too many complicated hobbies. And this seems like it would be a bit much, unless I just buy a premade bag.

1:05 It takes 2 weeks inside the fitting room for it to fruit? Is that normal, how long do oysters normally take because i'm having some trouble with mine, they take like 3 weeks and i'm not sure if it's because the conditions are not optimal or it's normal.
You shouldn't set petri lids face down like that, always face up. The steel surface is merely disinfected, whereas filtered air is sterile.
No sectoring?
When your making those LM transfers wouldn't they contain multiple spores? So it would be far from an isolate.
Please explain - It is my understanding as the spores are dispersed, they will germinate and the sectoring only takes place after the hyphae interact with another spore isolate. These germinated colonies of hyphae are isolated onto a petri dish before they have a chance to pair - thus creating isolates. Sectoring will happen in the next process
@@FreshfromtheFarmFungi I understand. I guess that wouldn't be necessary if you were able to limit the amount of spores gathered on the plate prior, like if say 1-5 all fell onto their own plate? And even then there's no way 1-5 would only contain just 1 spore, so they would inherently mate immediately after germination?
yes there is this error, but the spores that hit the outside have a greater probability of being single spores - only the next step will determine if they are compatible or not - possibly letting them go on the first plate and taking sectors after they grow out would be just as easy or easier - Thanks for the idea! In the end I just want a better lions mane culture

Do you have to do anything in particular to trigger the fruit body to drop spores? Or does it just drop them by default after harvesting? I'm a total noob but am soaking up so much info from your channel. Thanks so much!
they drop their spores based on maturity- you want to pick them just before they drop them

My lions mane on agar doesn’t look like my turkey tail or other species . It’s such a thin little layer on agar and doesn’t look like other peoples . Why does it do that ?? It’s normal ?
yes it’s normally thin and wispy until it fruits out
Awesome dude. I think the second flush should fruiting should provide better genetics, it is just my theory, make use to keep track witch bag fruits the quickest and then compere first fruting with the second flush. So the theory is that genetics are more established in the 2nd fruting compere to first, there fore creating stronger genetic code, what do you think brother?
It might be possible - I think epigenetics can play a role in this if the mushroom is more developed later on - the only way to verify would be genetic tests

About how long do you keep each dish under the fruit to make sure to not get too many spores

What temperature should I be pouring agar? I poured at 160 degrees the other day and had condensation. The next day I had contamination. Should I be avoiding condensation and would that have caused the contamination?
I pour mine around 160 all the time - I leave the plates in front of the flow hood for a day or 2 after to allow the condensation to evaporate. The contamination could have come from anywhere - this is where troubleshooting begins. Most likely it is an error on the technician. If you sterilize agar in a jar, leave it closed and it contams, then there is a problem with your procedure, if not, there is a problem with your pouring.
You are not worried that most of the spores will be incompatible with each other i.e. sister spores and therefore not be able to fuse? I would be looking at using spores from 2 different lions main strains
maybe but it’s a lot simpler - going for the “quick and dirty” approach- shortcuts usually are not as precise
That's not usually a concern, is it? The sheer amount of genetic variation available when you're talking about millions of haploid spores seems like it would take care of any incompatibility among spores.
You can't clone them by tissue?
you can - but this was for breeding purposes
did you get a faster Lions mane yet? the one I use is 10-12 inc and 9-10 day fruiting.
I have 3 different strains - I like the one I have been growing because of the density but that is probably why it takes a little longer

You can't see that it's a single spore isolate, you need a microscope. There could easily be two spores there and you can't tell with the naked eye. It's LIKELY it's a single spore, but you don't know.
How can you be sure that the spores are isolated?
with DNA testing - this is not a perfect process but a lot better than the previous method I was using for lions mane - another approach could be to let them breed on the plate, wait until there are clearly defined phenotypes and select them out when they have isolated on the original plate - the issue with this is that you would never obtain the haploid progenator. Moving forward there could be mated pairs that I consider “isolates” but that will reveal itself in the next step when it doesn’t mate with the other pairing. Hope that makes sense - shortcuts are never perfect
