- Hello I'm Dr. Shaifali Mathur (Ph.D. Biotechnology). I make videos on concepts in easy and simple way for beginner's in Biotechnology.
In this video I have explained Chromosome walking technique in easy Hindi Language for learning and making notes on the topic. Easy to learn and understand for beginners.
Here in Hybridization of probe step we perform colony Hybridizarion technique used for screening of genomic library.
In this step the bacterial colony is transfered to nylon membrane and denatured to expose the naked DNA . Then this membrane is soaked in a solution containing the labelled probe. Then washing is performed to remove nonspecific binding. The nylon sheet is sandwitched on Xray film. The place where Probe is bound is analyzed through autoradiograph and compared with master colony to find the exact segment of genome cloned in that colony.

Chromosome walking is not a sequencing technique.Only restriction mapping is performed to identify subsequent marker gene

Ma'am ... There's a question in my exam about two evidences which proves that dna replication is semi conservative.
First exp was meseleson and Stahl exp but which one is the other ??? Pls reply .
Other one is experiments of Jhon Carins that proposed theta model (formation of theta shape in other bacterium) of DNA replication.
One more is During the 1960s, Reiji and Tsuneko Okazaki conducted experiments in E. Coli to prove DNA replication is semi discontinous

Oh yes! Taylor experiment(1958) is more appropriate answer. as it proves that DNA replication is semiconservative.
