    Dear Johanna, thank you so much for the video. For instance, I would like to select 60x120 micrometer ROI area. How could I perform it? Thank you.

      Hi @merveemen3838. Select the rectangular tool, draw a box on the region you want to measure, then go to Edit > Selection > Specify. Fill out the width and height with the area you want.

    @johanna.m.dela-cru, can we measure the intensity of the fluorescence in a video, like how with time the intensity increases? is there any way to do that in imageJ?

    How does one measure the size of the ROI? What are the units of the measurement??? It just says area and a number; what does the number mean and what does mean, min and max mean?

      The size of the ROI depends on the thresholded pixels. If your image is spatially calibrated (e.g. in microns), the area measured will be in microns. If your image is not calibrated (meaning in pixels), the area measurement will also be in pixels.
      Mean is the average intensity of the ROI. Min is the lowest intensity found in that ROI and Max is the highest intensity value found in that same ROI.
      All other measurements that you want to extract from your ROIs can be set under 'Set Measurements'.

    Very informative, thank you! I am trying to combine multiple ROIs in the ROI manager (i.e. opposite of the split function) so that when I use the "measure" function I can obtain one measurement consisting of the total area. Is this possible?

      Hello @emanharrison. The ROI Manager has a merge function. Highlight the ROIs that you want to combine; go to More, then Merge.

      @@johanna.m.dela-cruz Thanks for the reply but when I select the ROIs then go to "More" in the ROI Manager, there is no "Merge" function... I have the latest 1.53t version of ImageJ.

      Ooops, sorry. It’s not merge - use “or (combine)” instead. You will need to 'Add' the combined ROIs to the ROI list.

    Hi, i am New in imagej and Hacer a question if could you answer please?
    İ am trying on one image that selecting multiple rois and want to for each roi apply crop, duplicate , add gray,Binary.......and skeletonise. Making it step bu step so bored. İ tried to make a macro but i failured. Can you help me please how can i handle. Thanks

      Hello @captnwho. You could probably do all those steps by using a macro. There are several macro experts in the image.sc forum (forum.image.sc).
