My anecdotal story: I'm a 60 year old male in Palm Springs, CA. I started taking NR(NAD precursor) about 6 months ago. I dropped from 240 pounds to 190 pounds and have kept the weight down, but bounce up and down with about an 5 pound variation. I believe the benefit of NR was to stimulate my exercise program which is basically 1 hr walk in the morning, 1 hr walk in the evening, plus intermittent gym based light weight lifting on machines. I have also tried variants of fasting, but have found that a 5 day water fast consuming only water and green tea works extremely well from a psychology point of view. Once I get past the first 24 hr fast, then the next 4 days are much easier because my appetite is suppressed. I'm taking NR during the fast. Also, started watching Dr. Stanfield about the same time I started the NR regimen(began with 300 mg per day, but now take 600 mg per day). The youtube support from Brad and others is invaluable. Thank you for helping everyone.

57:04 "Nobody has shown yet that you can do partial reprogramming in an old mouse and actually restore youthful function...", except David Sinclair's lab did a year ago.....the study was titled "Reprogramming to recover youthful epigenetic information and restore vision" and it was published in Nature. The lead author was Yuancheng Lu. How could he have missed that?
"...or increase lifespan" - In order to do this you would have to deliver the gene therapy to an old mouse's failing internal organs. The concept is pretty sound, however, the required biological engineering to accomplish it remains to be worked out. That is why Sinclair's lab started with the eye, because it is a complex organ that can be accessed without cutting open the animal. I have to wonder, for a guy whose next big study is the effect of intermittent fasting on canines, why he feels the need to throw so much shade at people's work.

A plant-based Mediterranean-type diet comes out at the top for good health, limiting protein intake and employing moderate IF (12-14 hours daily) to reduce mTOR, moderate alcohol consumption (1-2 glasses of red wine,) and moderate daily exercise - these are your best bets right now and dovetails with rapamycin research which looks promising.

    2. feeling physically sharp is very different from actually being physically sharp (low carb decreases work capacity, relative perceived intensity for the same intensity increase on low carb, motor unit recruitment decrease on low/lower carb ,which in turn lowers the potency of any working set done in the gym for a given movement)

Rapamycin half life depends on type. Sirolimus half life is longer, ~60 hrs. Also depends if we are referring to terminal half life (~80hrs) or elimination half life, as Sirolimus' curve is parabolic as it drops sharply, at least pertaining to blood concentration likely due to cell entry. Everolimus has a much shorter half life.

1:15:37 An interesting comparison would be Rapamycin verses Metforman vs. 1-3 days of fasting. It could be that fasting provides the most benefit with AMPK activation (and with no side affects).

Glycans are a really interesting measure of biological ageing. In addition, it would be interesting to cross check with methylation. I'm in the process of checking this out on myself. I'll report back when I get the results.

Great and “ontnuchterend” (putting un back on the ground) are these words from Dr. Kaeberlein. A confident speaker.
I have spend 2 months taking in 500mg of Resveratrol, can’t say I became Superman.
Because I also started watching my diet and sugarintake, I feel better.
I also take NMN, 1 gram per day. I must say (anecdotal) that my endurance has improved by around 20% without doing any workouts. I know because I did weekly self assessments.
I have been watching RUclipsrs citing clinical human trials with NMN that did show improvements.
I am therefore puzzled by what Dr. Kauberlein says here.

There is several key points to this presentation however I will narrow it down to this question 2017 NIH paper, Towards natural mimetics of metformin and rapamycin. Does metformin, berberine still fall in the mtor agonist category?

Great discussion. I'm thinking of adding Rapamycin to my regimen. I'm already taking Metformin. Can the two be taken together safely? I'm seeing conflicting information, some saying it is safe some saying it isn't. Is anyone here taking both? Thanks in advance.

    NADA precursers:
    1. Has anybody duplicated Sinclair increasing mice lifespan by 25%?
    2. Has anybody done any tests--on skin, muscle tissue, bone tissue, blood cells--to see whether NR or NMN supplements are getting past the liver to these tissues. And, if not, what's the delay?
    3. Seems to me if we indeed can show NAD decreases with aging(any actual proof?), that #2 above would be easy to do.

My. Wife’s perimenopause was reversed rapidly with 3mg of Rapamycin (Sirolimus) per 10 days. Also, her HHT (a bleeding disorder) was ameliorated about 80%. For your study, seeing if women’s perimenopause could be reversed would be a quick and clear clinical endpoint.

@@PakistanIcecream000 No, it wasn’t prescribed to her. I read that Rapamycin may help with her HHT blood disorder, and I was already taking it for longevity and had some I’d ordered from India. So she started.

It's always alerting when c. elegans is used as a model organism for aging research. C. elegans has a programmed aging mechanism because it is evoltionary beneficial for them (limited number of descendants, very limited amount of resources, so it is beneficial to die when maxiaml number of descendants is reached). So it is realtively easy to slow down nematode's aging: all you need is to break the aging program (e.g. mutations in daf-2 and age-1 genes increase their lifespan 10-fold). That's why I always sceptical about results on c. elegans

Does rapamycin restore mtor sensitivity similar to keto for insulin resistance ? We always assume more is better but i feel the right amount at the right time and sufficient sensitivity would be wiser? Insulin is great but clearly we overuse it. Perhaps same for mtor?
