    thank you for your video. But I have a question: can I traceback according to the largest value, so it would be 2, 1, 0, 0, 1, 0 instead of 2, 1, 0, -1, 1, 0?

      But according to this method you cannot go to 0 after 2,1,0 as it's a match (G and G) so you have to go diagonally, which is 2,1,0,-1. Note: here you can't go according to the highest value if the nucleotide sequence matches. If it doesn't match then you can go for the highest value.

    But I wanna ask some question.
    Why do you add these digits - 2 , - 4 , - 6, - 8, - 10 & -2 , - 4, - 6 ,- 8 in
    First row and column respectively why do we need to add this zero
    These points are bit confusing please reply me..

      Because this is the 1st rule. Before filling up a matrix you have to fill the first row and column by adding the given score of a gap penalty (which is -2 here) with 0. And the first one will always have to be 0.
