Can more than one person lock a color at the same time? For example a double 6 is rolled with the 2 white dice and two players have 5 crossed off blue colors so decide to block off the blue row.
I think your instructions are wrong. The matching of the colored dice with the white is optional. Instructions state - Active player (but not the others) may then (but is not required to) add one of the white dice together with any one of the coloured dice.…
Further, in the instructions (unless this used to be a rule) when addressing penalties, the rules state “If, after the two actions, the active player doesn’t cross out at least one number, he must cross out one of the penalty boxes.” So if you DO use the white pair of dice, you don’t need to take a penalty if you choose NOT to use the color and white dice. The two actions being (1) using the pair of white dice or (2) using a color-white combination.
Let's just say on player 1's turn he unlocks the green lock and the green die is gone. When player 2 goes to do his turn does he still use the green die? Or does it get rid of everyone playing the game with the green die ..

True, it is optional but if you don't take one you have to take a forfeit. Four forfeits ends the game
