    Fasting is also very effective for restoring NAD levels. Yamanaka factors, too! Some of the many benefits of doing occasional extended fasting: Blood clotting is reduced and blood clots and arterial plaque are reabsorbed into the body. High blood pressure is lowered to normal levels very quickly while fasting. Fribrosis/scarring is reversed over time.
    Fasting increases nitric oxide release.
    Vitamin D plasma levels are increased as fasting improves metabolic health, and vitamin D in turn increases autophagy.
    Telomeres are lengthend and fasting also increases anti-aging Yamanaka factors.
    Fasting stimulates phagocytosis, the ingestion of bacteria, plaques and viruses by the immune system. It will also remove any 'foreign material' that is not supposed to be there.
    Reflexes and short term memory are increased.
    Fasts from 36-96 h actually INCREASE metabolic rate due to norepinephrine release!
    Thymus is regenerated, which suppresses aging and renews the immune system. The thymus also plays a vital role in fighting cancer.
    Weight loss from daily caloric restriction has 1/4 to 1/3 of the weight lost as lean tissue while many studies show fat loss from 36 h fasts without losing any lean tissue! The hunger hormone ghrelin also lowers with extended fasting and rises from dieting.
    Blood sugar and insulin are lowered, allowing white blood cells to move more freely throughout the body and do their job. Ideal blood sugar is around 80. Some viruses activate glycolosis (the release of sugar in the body) and clinically it has been shown that decreasing glucose metabolism in the body weakens the influenza virus.
    When you move out of MTOR your body shuts down the building blocks of the cell which are used to produce organelles and proteins. This means the mechanisms needed by viruses to replicate are by and large unavailable when you are in a deeply fasted state.
    What breaks a fast? Anything with protein or carbohydrates in it will break a fast, though if the amount is tiny you will go back into ketosis very quickly. Most teas and herbs are OK. Most supplements and meds will either break ketosis directly or contain a filler that will. Many medications are dangerous to take while fasting so you may have to talk to your dr. about discontinuing them during a fast.
    Does fasting lower testosterone? No, it raises it when the fast is broken by increasing lutenizing hormone. Fasting also increases insulin sensitivity, which helps with muscle building.
    Fasts of 36-96 will not affect short term female fertility or affect menstrual cycle. They also may increase long term fertility, especially in women with PCOS.
    The hormone Leptin is an immunomodulator that keeps the body from attacking itself and obesity causes leptin resistance. Fasting very quickly reduces leptin resistance and leptin levels and one day of fasting can cut your leptin levels in half and gets your immune system working properly again!
    Does the body preferentially prefer glucose as a fuel? No, except for brief periods of very intense exercise, your body mainly burns fats in the form of free fatty acids. Your brain also prefers to burn ketones at a rate of around 2.5 to 1 when they are available in equal quantity to glucose.
    Deep ketosis virtually eliminates chronic inflammation in the body. This can offset the life threatening symptoms of viral pneumonia which effectively kills you through inflammation. This also creates BHB ketones in your body, which help your immune system and anti-oxidative system, especially in the brain. When not in ketosis, the brain can only burn carbohydrate, which produces a great deal of damaging ROS the brain has to deal with.
    It increases mitochondrial function and repairs mitichondrial DNA, leading to improved ATP production and oxygen efficiency and thereby making cells better able to fight off infection. Increased mitochondrial function also has the added benefit of increasing your metabolism and cancer prevention!
    When you fast, this stimulates apoptosis in senescent or genetically damaged cells, which destroys them. Senescent cells are responsible for many of the effects of aging and are a root cause of the development of cancer.
    Fasting also releases BDNF and NGF in the blood which stimulates new nerve and brain cell growth. This can help a great deal with diseases like MS, peripheral neuropathy and Alzheimers.
    In fact, the biochemical regulator of BDNF production is beta-hydroxybutyrate, which is the same ketone the body produces to nourish the brain while fasting.
    Fasting also increases telomere length, negating some of the effects of aging at a cellular level.
    A fasting mimicking diet for 3-5 days in a row also provides many of the same benefits as water fasting. FMD usually has 200-800 calories, under 18 g of protein and extremely low carbs.
    Exogenous ketones can aid with fasting, making it easier in healthy people and allowing some people with specific issues to fast in spite of them without worrying as much about hypoglycemia.
    Children, pregnant or nursing women should not fast for periods longer than 16 hours. People with pancreatic tumors or certain forms of hypoglycemia generally cannot fast at all. Type 1 diabetics can also fast but it is more complicated and should be approached with caution as it could lead to ketoacidosis. Those with Addison's disease may also be unable to fast without liberal use of exogenous ketones, depending on severity. If you experience extreme symptoms of some kind, especially dizziness then simply break the fast and seek advice.

      Does this imply that a keto diet is best? I could never go keto as I enjoy a balanced diet and am a long distance swimmer (I need calories). Can you go into ‘ketosis’ after just one 36 hour fast? That’s about the longest I would ever voluntarily go. I’m a 64 year old woman, btw, just for perspective!

    Fascinating.
    I'm aged, have always been a physically active adult. I am dismayed at how quickly my energy fades in spite of eating a healthful diet, and being fit. If I had time, I'd be hitting the gym, riding bicycle. I've a huge landscaping project started... hard work.
    I run out of juice at about 2pm. I have to rest, nap, whatever.
    I am taking NMN, and it doesn't seem to help at all. This video explains why. Thanks.
    I've a lot of work to do, I'm a musician, craftsman, metal worker. I cannot waste time napping.
    I'll be looking up this product.
    I take a lot of supplements. I know they are not drugs, take time to work, and work slowly, at best, if noticeable at all. I'm in much better shape than my peers... many have passed from lifestyle choices, others too tired to get off the couch.

    2. Take cd38 inhibitor: apigenin.
    3. Take NAD recycling enzymes: NAMPT.

      you got it wrong. their product does NOT contain the enzyme NAMPT. One should not mess up the metabolism by taking enzymes directly.
      Next, the point is that NMN, NR are nt as effective, for several reasons. Here it is about NAM

    Very helpful!
    They want to take NMN and maybe NR off the market as a supplement. These are both precursors to NAD.
    If these are naturally occurring, they should remain as supplements.
    If David Sinclair wants to market his crystallized form of NMN as a drug, that should remain a separate issue.
    People will not be able to access something like this if it is prescription-based, and be able to enhance their own health.

    It would be nice if the company Nuchida could post their clinical findings. This is nice to have a mix of products but i am skeptical about the outcomes vs other products. Especially their product prices are quite high. thanks

      yeah seems it is cheaper to take NMN, The whole sell story have one flaw if you stop taking their 65Eu supplement the factory stops right. Same thing as taking NMN.

The more I watch, the more supplements I jot down in my spiral notebook. Quite a tall stack of spirals now. Think I'll be adding parsley to my salads more often.

      if you mix the ingredients by yourself, you end up with the same amunt. Consider that their product is high dosed, so you can calculate to take only 2 caps per day. and then it is quite affordable

    I was taking NAD for awhile..thenI started reading reports of it being dangerous to take for long period of time. I switched to taking Nicotinamide (also recommended by a RUclipsr). But I also take several other suplements since Im vegan (Vit D3 K2 Vitamin C and B12). Im 71, no serious illnesses and regularly get told I like mid 40's or 50's. Guess I'll keep doing what Im doing😅

      Thanks for your post. I heard about cancer with NAD so I am nervous too. Is there no risk with NMN? Have you noticed a difference? I learned we want the Methylated version of all Vits. including B6 and B12 should be sublingual too. Some people have a MTHFR gene so only sublingual will work so that is what I do. I also really hope you are taking Magnesium Glycinate before bed. It is essential for most. I have been taking it for many years.
      Try to avoid synthetic forms of vitamins made from petrochemicals. Read the labels. One trick I use is to compare to a really expensive premium brand. This way you can see what should be in it- and more importantly, what fillers and things should not be in it. Read the new book called YOUNG FOREVER- Dr. Hyman. It is excellent. At 71 seems you are doing many things right. Good for you. Also, do check your blood sugar levels twice, 2 days a week. This can help you adjust your diet to lower your sugar/carbs.
      I tried being a vegan- love the idea behind it but it did not work for me. I quickly got deficient in many things and felt terrible. Sad to say, I am no longer vegan. I really wish I could be. Even as a non-vegan I have trouble gaining weight. I did lose my gallbladder- not sure if that has anything to do with it but maybe. I probably do not digest fats too well- that is what it helps with.

      @@MOAB-UT Hi, there is no evidence that NAD+ causes cancer, in fact NAD+ levels and cancer risk are negatively correlated throughout life. Cancer risk rises as NAD+ levels decrease with age, both exponentially. What experiments do show is that NAD+ production can be targeted as an Achilles heel in cancer cells. If you cut off the NAD+ supply cancer cells die. The same applies to glucose and oxygen. Cancer cells are heavily dependent on a supply of critical molecules and removing the supply locally kills cancer cells, but this does not mean glucose, oxygen or NAD+ cause cancer, nor is it a sensible approach to restrict any of these critical molecules.

      @@chidude Thanks and no. I think it has more to do with being used as an amino acid but only during times when your body is stressed. For digestion I've taken a digestive enzyme in the past because I lack a gallbladder. But thanks for offering your thoughts. I also eat plenty of meat so I think I get enough from natural sources which is usually better. Nutrition has also been my hobby for the last decade.

    I started using 500 mg nmn per day since 30 days ago and it literly changed my life. I have more energy, my brain fog is gone. Also i was always tired in the morning and lit up in the evening.       Methylation is a critical biochemical process that occurs in the cell when a methyl group (consisting of one carbon atom and three hydrogen atoms) is transferred from one molecule to another to help that molecule perform its function.
      In a nutshell, because of the methylation cycle, methyl groups are required for many crucial processes in our bodies such as producing neurotransmitters and antioxidants, epigenetics (how DNA expresses itself), regulating cardiovascular and nervous system activity, energy production, and heavy metal detoxification. Our bodies have a reserve of methyl groups that it uses whenever it requires methylation for cellular processes and functions.

      @@cinystarr4657 look out for the rutin and troxrutin. These are plant compounds which help to activate\enhance availability of NAMPT. you get it alone as homeopathic preparation, or you can find it in skin of peach, or in the plant she describes. r-ALA is very helpful as well.
      In sum their product is outstanding, and i did a lot of research. If you try to mix compounds from your own sources yu end up paying much more.
      That said, you probably do not need 3x2 pills per day. for me 1..2 is already enough, it is very effective

      @@petermalmgren1207 is not precisely the same. Niacin is converted into NAD+, while NAM is created FROM NAD+. The latter is relying on a proper salvage pathway cycle, hence the preferable intervention, while niacin can disturb a range of feedback mechanisms when taken in higher dose. Admittedly it is a bit cheaper. Both approaches share the need to co-supplement with TMG

    That is, is D-Ribose a possible component in your mix?
    My first scan of that report was pretty inconclusive. I don't think they sepatated the effect of the two substances, so I couldnt say if all effect was from Nicotinamide or not.
    And it was just a short pre-study.
    Furtermore, I wonder if D-Ribose is just one molecule, or a family.
    They seems to think it does something, though.

    Does this account for the possibility that an increase in NAD+ might also enhance the growth of cancer tumors? There's a cost involved with increasing NAD+ (according to other doctors in this field) and I'm not sure the public are aware of it.

      NAD+ levels and cancer risk are actually strongly negatively correlated throughout life. Cancer risk rises as NAD+ levels decrease with age, both exponentially. There is no evidence that NAD+ causes cancer. What experiments do show is that NAD+ production can be targeted as an Achilles heel in cancer cells. If you cut off the NAD+ supply cancer cells die. The same applies to glucose and oxygen. Cancer cells are heavily dependent on a supply of critical molecules and removing the supply locally kills cancer cells. This is one of many alterations in a cancer cell that evolve to confer a survival advantage. Which is why taking NAD+ supplements isn't recommended for those with active cancers.

    Have you seen the study that says that microdosing pure nicotine can stimulate/activate NAMPT? I know nicotine has very negative associations from tobacco, but in its pure form it looks really interesting.     6:20 CD38 doing nothing but draining NAD, "unnecessarily" , while also being part of an inflammatory response, doesn't make any sense. Instead of inhibiting this enzyme, couldn't we somehow help it deal with the excess Ca++ it tries to remove?

      DC38 is an enzyme used by the immune system for pathogen fighting, so you don't want to lower its level directly. Fasting for example, kills off the pathogens so less cd38 is triggered, but trying to fight cd38 like some of these yt channels suggest is not going to be helpful but harmful.

My half-way educated guess would be that there's something aging the ER and/or there's something off with magnesium signaling/availability, leading to CD38 going rogue. Magnesium ions act as cofactors for several enzymes involved in calcium signaling, including those that synthesize and degrade calcium messengers such as cyclic ADP-ribose (cADPR) and inositol 1,4,5-trisphosphate (IP3). Common symptoms of NAD+ deficiency are decreased physical and mental energy, brain fog, disrupted sleep, reduced ability to recover from illness and loss of strength.       I would say, the activation of the AMPK pathway through r-ALA is an opponent of cancerogenic processes.
      However, if you messed up your metabolism by eating to much sugar, or high GI carbs, snacking all the time and such provoking a conitnuus hyperinsuiinemic state, and never doing a 20+ hours fast, THEN you should NOT pour a pure energy booster into the system.
      afaik, the cancerogenic hypothesis came from in vitro experiments. Also note, that many tests are made with mice and rats, and those have a natural tendency to die from cancer in old age. ...unless us, IFF... see above

      I think you are suggesting that AMPK activation via stressors (hormesis) like exercise is a good thing- right? If so, I agree.
      Even more than just sugar, I think the real issue is inflammation. When we consume seed oils, processed foods, fail to exercise, stress out and lack sleep, our bodies are primed for trouble. Then consuming sugar is pouring fuel on an existing fire. There are some younger athletes I follow like Mikey Musumeci. He is a machine- virtually unbeatable. His diet is absolute garbage consisteing mostly of heavy carbs Pizza and Pasta. He is proud of that. Yet why is he so dominant? I guess it can take a while to catch up and he is very active so he gets away with it...for now. There are many factors to the health and wellness puzzle. In addition to running and working out, I have started to eat 3 cloves of raw garlic and raw red onion with Avocado and EVOO daily. I put them on an Outer Isle Cauliflower round (instead of bread.) Only two net carbs. You can grab them at Costco.

    The salvage pathway "simply doesn't work as well". Now, those are words even non-scientists should refrain from using.
    A few years back we'd say, "efficient cellular respiration, simply doesn't work as well with age", which led to sketching the importance of NAD in age reversal.
    "Diminishing NAD salvage pathway", should be the answerable issue. Otherwise, desperately pumping the freshest air in a flat tire, simply may not work.
    Instead, looking into how the original blocks - which do create life - could be ad infinitum optimized, might offer a clue. Answer hovers over "not destroying", and "creating". Be happy.

    You say that CD38 drains NAD out of the cells "unnecessarily." but is that actually true? CD38 is an important part of the immune system and perhaps it needs more NAD to function or be added for a NECESSARY immune response, particular in older individuals who have an involuted thymus and therefore fewer T cells, each of which may need more CD38 to be placed on it's cell membrane.

      As you say CD38 does have an important role in immune function, however during aging expression of CD38 increases greatly. This wastes large quantities of NAD+ (CD38 is actually the largest mammalian consumer of NAD+ wasting around 100 molecules of NAD+ for every reaction is performs) and promotes further chronic inflammation and cellular senescence. CD38 also exerts dominance over other beneficial NAD+ consumers such as the sirtuins meaning these critical pathways are left without sufficient NAD+. In this manner extremely high levels of CD38 are not beneficial. Studies have shown that inhibiting CD38 even slightly can boost NAD+ levels, which is why Nuchido TIME+ contains apigenin.

    Dr Nichola Conlon talks about addressing the route causes of declining NAD+ yet her suggested solution is supplements to boost the NAMPT level, provide NAD+ precursors, and reduce the NAD+ destroying enzyme, CD38. While not doubting the efficacy of this combination, I can't see how it addresses the route causes. After all, we don't need these supplements when we're younger.
    Wouldn't addressing the route causes mean improving the health and increasing the number of mitochondria, where the Krebs cycle takes place, and reducing the number of CD38 excreting cells, if there are too many of them? Ginger, for example, has been shown to increase mitochondrial size and mitochondrial biogenesis and can be purchased very cheaply and conveniently as ground ginger. Ashwagandha has also been shown to improve mitochondrial health and function.

    .... but if thats true what u said, the inflammation is the cause of everything, or before that, the immune system. Wouldnt it b better to either restore the immune system or get rid of these inflammations for example by just using curcumin?

      I theorize that we are so inflamed because we eat too many plants.
      Switching to a meat based diet drastically reduced my inflammation.
