    If we have inserted the gene of interest in tetracycline gene then the bacteria will not grow upon treatment with tetracycline then how can we collect it if it dies ????

    Dont all the bacteries have antibiotik resistance genes how do we differentiate the ones that are inserted or not

      No, often bacteria with resistance plasmid is resistant to some antibiotics. We differentiate by replica plating. Only bacteria which is transformed with vector can live in ampicillin or tetracycline containing medium. Thank you
