Leishman is used becoz it contains both acidic n basic dye , acidic is eosin n basic is methylene blue .
In leishman stain preparation we add eosin n methylene blue dye in acetone free ethanol . the eosin part of leishman stain help stain basic cytoplamic component while basic methyleneblue part help stain acidic nuclear part . acetone free ethanol is used as acetone is a good lipid solvent n it may result in disruption of cell membrane so acetone free is used
