Important to note that APOE has different types, APOE e4 being the worst and most highly associated with Alzheimer’s disease. Meanwhile APOE e2 is considered to be beneficial in protecting against Alzheimer’s because it is much better at clearing out amyloid plaques.
The AICD fragment cleaved from the amyloidogenic pathway functions as a transcriptional factor for the expression of Ca2+ related genes and other genes resulting ultimately in neurodegeneration. Also the AICD in the non-amyloidogenic pathway is also simply degraded in some cases and does not act as a transcriptional regulator.
im not a medical guy but something is unclear about an alzheimers brain using b-secretase instead of a-secretase. why is the wrong type of secretase there ?
does the Abeta40/42 itself cause the a-secterase to switch to the wrong type in a runaway chain reaction somewhat similar to mad cow disease BSE? or is b-secretase a completely different enzyme?

question, does this mean that normally beta secretase is not involved at all? also, if so then how come it suddenly becomes involved in a disease brain?
I am not positive, but in my research I figured out that there is genes for how APP is processed on the 21st chroosome- which may explain why AB is related to downs syndrome (21st chromosome deletion).
