  • David Sinclair PhD is a biologist and Professor of Genetics at Harvard Medical School, co-director of the Paul F. Glenn Center for the Biological Mechanisms of Aging and author of the forthcoming book "Lifespan: The Revolutionary Science of Why We Age -- and Why We Don't Have To".

    I am 86 , retired, healthy also have some aches and pains, mostly from a bad auto accident .I am finding out that I am tired of being retired, it is boring, I want to start a new adventure , a new carrier, I am starting to do what Dr. Sinclair is talking about , I think there are a lot of people out there who would be bored and want to help to make the world a better place , to feel useful again, if they could reverse their aging and be young again.

    I am 70 and have practiced only eating when your hungry missing meals drinking apple cider vinegar etc I am also a long time meditator so I look in my 50s that’s others perception. So it works!

    Really interesting from 1hr 30 onwards when you got onto challenges around the philosophical and practical challenges of population increase and so on. David Sinclair more impresses with his fully rounded perspective on the work he is doing and likely impact.

    I have been watching Dr. Sinclair's interview and this is the best so far. Very informative conversation. His book is also very educative. I love that book. In there, there's no point increasing lifespan without healthspan. The goal should be to live longer while staying young and healthy. Sinclair believes that in the foreseeable future, it's possible for humans to live beyond 120 or even 150, without the frailty, diseases and disability we now associate with old age. I hate aging and the illnesses it brings over time. This is why I am now taking NMN supplement by Vitruvin constantly.

    No matter how long you live it's trained mindfulness which really makes each moment come alive and more enjoyable. For that Thich Nhat Hanh is the master to learn from.
    My takeaway is to up my lvl of exercise and to keep improving my 8-year experience with a healthy and fresh, highly varied plant based diet. Other than that I will try to get more hungry more often. I will keep going with my cold showers and my intermittent fasting regimen and will try more regular sauna.

    I love every interview of Dr. David ever since he started discussing longevity. I have also acquired his book "Why We Age and Why We Don't Have To." That book is eye-opening and educative. In there, there's no point increasing lifespan without health span. The goal should be to live longer while staying young and healthy. Sinclair believes that in the foreseeable future, it's possible for humans to live beyond 120 or even 150, without the frailty, diseases and disability we now associate with old age.     Doing hard PHYSICAL work(I'm 65, a window cleaner), not playing with a computer mousse all day, I believe helps too.

    I have been on NMN and Resp. with vegan diet and B12, D3, PQQ supplementation for about 15 months. I cut out alcohol for that 15 months and exercise a lot. My wife and I did the Dr. Longo fast once back in April 2019. Last year was the best year of my life for training and general health. All my markers are good for a 60 year old. I went off everything for one month (Dec 2019) after completing Ironman Cozumel and felt like (I used to). Now back on the program and look forward to getting back to training.

      Thorne Research Labs has one of the better resveratol supplements available. It's very pure. Third party tested and noticeable in its effects after a couple weeks of taking it. It's also combined with the Quercetin that Dr. Sinclair also praised. I'm not sure if he backs this company or not but it's a very reputable company backed by other brilliant minds su h as Dr. Rhonda Patrick who's also been on RR multiple times.

    In Huxley's Brave New World they talked about about shortening the disease phase at the end of Life, something like people don't wilt with old age they snap and die quickly when the time comes

    Great work Dr. Sinclair! I was very interested to hear you are working on iPSCs like the Belmonte group. If selling a house were not so expensive I would apply to the PhD program there and come help! My MS in biochem. is pretty rusty (1988) but I've been thinking of helping out on the aging/health-span work as my 5th career (20 yrs. USAF/pilot, taught chemistry/biology many years) before I hit age 76 next summer! I'm 6 hrs. SW in Reading, Pa. w/ no comparable school in the area.

    What a FANTASTIC interview and discussion! I always thought it was ludicrous that we get to an age where we are wise and experienced, and then body starts to fall apart. Crazy! I had no idea that an aging drugs and research were so close at hand. I'm 64 and looking forward to the drugs, as long as they are safe. Meanwhile I should eat less and exercise more.
    Also I see that pet lifespan will also be increased, making dog owning a lot less stressful!
    Maybe when we know we can live a couple hundred years we will be much more careful about preserving and healing
    our beautiful Earth environment. And also be much more responsible about procreating.

      Just unfortunate that he mentions eating a steak is the worst thing you could do..ONLY on a vegans podcast. He never mentioned it on Joe Rogan's podcast or others I have heard even when he is specifically asked what they should do to stay looking young.

    Adam in the bible died at the age of 930 years old and his descendants died at the age of 800 to 700 to 600, etc and Joseph died at the age of 110 years old. We will be lucky to have Dr. Sinclair help us live up to 200 years old with this poluted and processed food world.

    So at this point the question is: How do we determine the levels of "healthy" stress (hormesis). If we overdo this, we will do more bad than good.

      It is a vital question. To use the example he discussed, fasting decreases fertility in women because hormones that regulate ovulation and probably other functions are incredibly sensitive to energy intake. On the other hand, NMN that is activated by exercise or fasting also has the effect of erasing symptoms of menopause and restoring regular monthly cycles, potentially making it possible for older women to become pregnant again. All we can do for the time being is regular blood testing to track biomarkers that may be thrown out of range by any of these techniques or supplements.

      betzi b yes, I do. As long as you ingest healthy fats and nutrition when you do eat, then you should be fine.

    My mom lived to be 93 years old. For the last two or three years she would sometimes say "We're living too long." She was in a fair amount of pain and discomfort most of the time from arthritis and allergies.

    If I could live to two hundred I would have gone back to college to study neurology and bioinformatics. Being 65 that kind of investment of time and money is just not feasible.

    I have pretty much lived this way for the last 35 years without really knowing it. Vegetarian, relative caloric restriction with a lot of aerobic exercise in extreme temperatures etc....Id be curious to be tested by Dr. Sinclair to see what if anything this has done for my health.....

      There is really no secret as far as what Ive done over the years. I run and cycle up to two hours total most days, sometimes more sometimes less. I typically only eat two meals a day other than say a lowfat yogurt and maybe a piece of fruit for "lunch". Im not vegan, I do have milk products on occasion but never any meat, fish, poultry etc...My breakfast is invariably a large bowl of oatmeal with maybe a few walnuts, some blueberries and brown sugar made with water. Dinner is usually some sort of vegetable concoction and maybe some bread of some sort. I do have a bit of a sweet tooth so I indulge in desert in the evenings. I burn a lot of calories through endurance training so I think the sweets help to fuel that. Not that it makes a difference but Im not a caffeine person so little if any coffee, tea or the like. Mostly only drink mineral water. As Ive gotten older I do take a multivitamin as a precaution.

    One interesting thing a friend of mine mentioned when i told him about all that stuff in relations to not only expanding the life-span but the health-span as well is: All the years of struggle we currently have to go through, the period of morbidity, getting hit by one desease after another, getting weaker and weaker and so on actually prepare us for death. At some point we just die out of resignation, no longer able and or willing to keep up the fight and we actually feel relieved of letting go and saying goodbye, knowing its for the best, its time, its all good.
    As David mentioned, by pushing the tail end of the vitality curve as far out as possible and when you are 130 you get sick for 1 week and you die would most likely result in people not getting confronted with their own mortality that much at all, clinging to life and their vitality and actually getting surprised of things ending so sudden. Not being able to make their peace with it.
    Our life-span increased by a lot and i guess its fair to say that our health-span couldn't keep up and therefor increased the period of morbidity. So shortening it would be surely a good thing. But shrinking it down to a week seems a bit too much and would result in other problems, as mentioned.

    I can appreciate this discussion, but the whole pharma route makes me uncomfortable. I can only imagine what this treatment would cost. Maybe he is right, in time it may be cheaper just like air travel.
    I understand he is doing a good thing for science. Such a huge topic for me to synthesize...living to 150.

    Dr. Sinclair, you are on the right track as it is the will of our Wonderful Creator to appreciate Creation and His awesomeness.
    Sir, evolution is NOT scientific, and we ate not animals; that is why we are conscious of death. Medical Science has it's root firmly established in Genesis chapter 2 of the Holy Bible. Our Wonderful Creator put Adam into a deep sleep, opened him up, took a rib to form a woman, Eve, for a wife. Hence, marriage between man and a woman is the foundation for a family!!! I respect the fact that some people feel/prefer to be different.
    DNA is a complete and the most complex form of program. It contains diverse forms of human beings in accordance with their environmental needs, which is adaptation. Adaptation is NOT evolution! DNA has the ability to repair itself.

    How about this: Aging is not a disease but a state of enhanced disease processes. Even if we live to 200, we're still going to die. I like the idea of thinking in terms of delaying disease processes rather than seeing aging as a disease and death as its outcome. We're still going to die, even if we double our lifespan tomorrow.

      Maybe. But we could still have a few millennia to decide. The anti aging field is barely 25 years old and Dr. Sinclair is already talking about living to 1000.

    Can't agree about forgetting about death from 6 onward. As your brain develops as a teen I certainly started to think about all kinds of questions like if there's a god and what happens after you die. Many people just accept there is a heaven which then allows them to stop thinking about death and get on with living which has it's advantages. Now we can hope that we can live an extremely long time and not worry about dying in that way and get on with our lives (although of course doing all the things required to increase your probability of long life, but these things should be habit, not conscious acts)

    the idea that epigenetic marks are lost upon DNA repair is excellent and the research looks great. One simple way to think about it all is that it's all about dynamic genomic integrity, and who needs NMN (or NR)?? -> if you can get B vitamins (niacin, remember is B3), have enough Magnesium (just the simple salt is fine) and anti-oxidants, and move a bit... just imho! ageing well for all is a great goal (not just looking for new things to market to the masses)! Take care!

    I want to live forever. I stopped smoking, limit sugar to less than 6G per day, consume NO bread or grains. Lots of nuts, fish, collagen and vit d paired with two miles of walk/run every morning. I just started resveritrol and started exercising in the cold. I'm noticing some anti aging already. That stubborn fat below the jaw is going away along with wrincles. I think my knee is also rebuilding itself 5 years after my ACL replacement surgery. I was able to RUN down a wet mountain trail the other day and I felt like a kid. No pain or soreness at all. I wish I could make people feel what it's like to approach their genetic potential (which is always greater than you would expect).
    1. No to low sugar
    2. No bread period
    3. Intermittent fasting
    4. Eat oily fish like sardines
    and stay away from strachy veg and legumes. (They cause bloating and inflammation)
    5. Walk/run every day within an hr of waking for at least 30min.
    6. Take good supplements

    I’ve not yet come across this question .. Has David Sinclair tested himself daily to monitor his level of NAD when using metformin, Resveratrol, and NMN? It would be good to know if one dosage kept his levels up for just only the day or a few days.. obviously everyone’s chemistry is different but it would still be a good measure to know. Thanks!!

      From watching previous and newer videos with Dr. Sinclair, he's been taking NR or NMN for ten years now and his 23 and Me test revealed that he biological age was in his late 30's or so if I recall correctly. The $64K dollar question is, how old are those mice now? How long did they live? Especially the treadmill mice!

    They touch on testosterone around the 1:23 mark and I am curious to hear more. He says increasing your testosterone could have a negative affect on longevity. As we age our testosterone levels go down, but with NMN he says his blood chemistry shows he is decades younger. Does the blood chemistry showing he is younger include elevated levels of testosterone? The studies in mice show that with NMN older mice can have the strength of younger mice. What accounts for this increased strength, are their muscles larger, more efficient, less fatigue?

      Perhaps it is increasing ATP in cells , so more energy is available. My great uncle, was a tall lean guy, with baseballs for biceps. He was not big, but wiry , he was an active logger well into his late 60"s and he was ripped.

      The process basically starts with the etabolism of fatty acids and glucose which requires NAD+ which is in turn necessary for the production of skeletal muscle to enable posture, breathing, and locomotion etc. So it is a major player in skeletal muscle development, regeneration, aging, and disease. I'm not exactly sure how testosterone augments strength in the beginning but comes back to bite later but the vast majority of studies indicate that lower NAD+ levels are harmful for muscle health and higher NAD+ levels increase muscle health.

    Sounds like great stuff. Lots of risk. Don't know if David is all he says he is or knows all he says he knows or how much of it is true but it seems to all line up with my own superficial look-see over the past few years. Again, giving I don't know him, I like his attitude and his humility. A little worried he is a little presumptuous. In this case it may be warranted :-) Usually isn't though. For example maybe some people would prefer to live to 60 with lots of muscles than to a hundred without them. Might even be the best thing for them...
    In this case - in a unique case like this - it seems that philosophical implications can affect humanity not unlike hard science discoveries, and so must be identified and then weighed carefully. Presumption eventually leads down blind paths.

      He was on Joe rogan and that podcast I believe he explains his background in the science far better then this.
      Second I agree him explaining the benefit/cost of certain diets and supplements is just opening an avenue for people to choice how they want there life to grow.
      I imagine a society where many will live longer as this data is harnessed into better information, then they will live longer but we all be different as a species.

    People it still hasn't been PROVEN that human life has been extended , David is still in the process to prove that , it has worked to some exstence in a mouse that had younger physical abilities but didn't show a comparison life span , NO facts that life is extended IN HUMANS BUT TIME WILL Tell !

    Scientists use jargon when it is not necessary to discuss simple terms all too often. Essentially niacin (B3) can convert to NAD and positively influence a bit of juvenescence in the cells. As I understand it, tryptophan can also convert a bit into NAD.
    By the way, niacin improves blood pressure and cholesterol. Do your own research.

      I don't recall Dr. Sinclair ever criticizing Niacin and it may be an original NAD+ precursor but it doesn't have a longevity effect he is looking for (at least in mice). There should be more in his book but there are real differences that aren't a matter of terminology.

      @@Elayman1 Hi there. I was not suggesting that he criticized Niacin. He simply never used the term, when in some instances it can be interchangeable. I can attest to seeing an effect with Niacin in a less clinical manner. Some other research has highlighted its benefits. I combine its use with three other natural products. I was born in 1967. Melanin is not always a factor. Lifestyle is.

    My naturopath suggested I do NAD iv once a month ... still trying to wrap my head around this stuff ... but if anyone is interested in that... they charge $300 for that ... one round... slow drip ... I may switch to oral supplementation, but will likely do another three or four iv’s at least ... it is more expensive than the other iv treatments which usually run around 175... or less ... I did it once so far ... I was unwell at the time, fatigued, recovering from a gallbladder attack, I’m anemic, super low on D and have a large benign tumor... all I can say is that NAD iv revived me.

      Cassandra Decosta while I don’t know how much more advantageous it is to do the iv version, and it may not be all that more advantageous... but, often iv’s are superior, probably not always though... and yes, the cost of the iv is quite prohibitive... I have considered going with supplementation ... I still feel like I need to understand more... and like I said, I will probably only do the iv for a short time... my ND recommended it once a month for now... I did not ask him how long... but I can manage the cost right now, so I will probably do the iv another 3 to 5 times and do plan to switch to a supplement strategy from there

      @Chew Chew Train but the carnivore diet helped so much when I was having major stomach issues and so many people have amazing results on it so I'm perplexed. I cant eat lectins or anti-nutrients from plants.

      Studies have proven that high carnivore diet is linked with more cancer. I believe Dr's Longo and Fung recommend plant based with small amounts of fish (best) a couple of times a week. More protein if you are a senior

    Good discussion. One clarification in your Dad's Story on the reason his employment services were requested for so long. It is not because Anchorage or Alaska has an enormous Native population and they value wisdom of elders. Both are true however Alaskans value anyone who has many year experience working in the Alaskan Environment, particularly Engineering and construction. I know first hand. My Grandparents came to Alaska in 52 when it was a Territory. They both African American lived to 99 and 97 as well as all there siblings into there 90s and mother 100. All worked into there 90s. A good friend of the family owned a construction company and buildt perhaps 10% of the city. He is 86 and still is called for advice and work by non native Alaskans who are the majority of the population. I have lived all my life in the state, while travelling to 33 countries and working few years abroad. My family's key to long life. Work, have a purpose and eat clean nothing processed nor excessively.
