American Gene Technologies' AGT103-T is a therapy being studied as " functional" cure, with means the virus is not completely eradicated from a person's body.
On February 20, 2023, the 53-year old man in Germany (named the "Dusseldorf Patient", documented as the *5th person totally cured of HIV* by receiving a stem cell from a doner with a *CCR5Δ32 mutation* , which blocks HIV from infecting the body was diagnosed with HIV in 2008, and after their HIV diagnosis, was placed on antiretroviral therapy (ART) which suppressed the viral load within their system.
The 53-year old patient enrolled in the University Hospital Dusseldorf IciStem program, which explores potential HIV cures requiring stem cell transplants.
This 53-year old patient was diagnosed with acute myeloid leukemia about three years after his HIV diagnosis. Remission of the cancer was achieved through initial rounds of chemotherapy, but was followed by a relapse shortly after.
During the 53-year old patient's treatment of his cancer, he received a stem cell transplant from a female donor with whom they matched, and the female donor had the CCR5Δ32 mutation. This stem cell transplant occurred about two years after the 53-year old patient's cancer diagnosis and five years after being diagnosed with HIV.
This stem cell transplantation, was a person with the CCR5Δ32 mutation is a complicated procedure, normally reserved for cancer patients, and comes with many risks, and is too risky to offer it as a option as an HIV cure for everyone.
With *AGT103-T* , developed by American Gene Technologies, currently in human clinical trials in the hope that this therapy can be a "functional" HIV cure and *EBT-101* , developed by Excision BioTherapeutics, also in human clinical trials, possibility AGT103-T & EBT-101could combined therapies in the possibility of having a " *sterilizing* " cure of HIV, much like stem cells transplantation from donors with the CCR5Δ32 mutation, but without the risk associated with it.
This of course is after both AGT103-T and EBT-101 have independently demonstrated for their safety and effectiveness before the possibility combing both therapies. AGT103-T and EBT-101 have completely different approaches.
*AGT103-T* grants HIV resistance to the T cells that are responsible for hunting HIV, allowing them do their job and fight HIV, like you would for any other virus. This includes continuously fighting the viral reservoir.
*EBT-101* , on the other hand, is a temporary pulse of CRISPR-associated gene expression that aims to cut out all HIV from the human genome, but since the CRISPR-associated genes cannot be continuously expressed, they need to cut of 100% of HIV proviruses during their expression window, otherwise HIV can just replicate and return.
American Gene Technologies will need to see updated data from both therapies to know how well both of them work and whether or not an AGT103-T+EBT-101 combination therapy would be warranted.
Both AGT103-T & EBT-101 are both in clinical trials, which usually takes between 2 and 5 years because each time a patient is infused, they are extensively monitored for side effects, efficiency and variations in their response to the therapy.
In regards to AGT103-T, in theory AGT103-T could be given a dose preventively, as a vaccine, but it would be significantly harder to make due to the lack of HIV specific TCR enrichment that are seen in PLHIV. Also, T cells which do not encounter their target will naturally reduce in number, so it's possible that AGT103-T would not be permanent in people who do not have HIV.
AGT103-T archives the same functional effects as delta 32 variant (CCR5Δ32), which some people are born with and when HIV tries to enter their cells, it can't grip onto the mutated CCR5 produced by these individuals. AGT103-T does deliver mRNA to prevent CCR5 from expressing on the cell surface, which makes cells resistant to HIV entry.

2. The amount of risk is a little high due to the risk of complications from the stem cells. worse case scenario the treatment can be more lethal than HIV/AIDS

That's a tough one because of how many forms of cancer there are and how cancer mutates throughout its stages. All in all, though, if cancer ever has a cure, the already overpopulated world will become even more overpopulated. Unfortunately, disease and various viruses is about the only population control this planet has for now. If we become too over populated, there won't be enough land for us all to live and we'll have to tear down all of the forest and eventually everything else will die. We'll soon follow.

There are resistant people. However, this “it is the black plague” hypothesis is not taken seriously at all by actual researchers for good reason. It is a made for the masses, Reader’s Digest level idea. As a matter of fact the research shows the opposite. From an abstract on the topic, “If anything, the gradient of Black Death mortality sloped in the opposite direction from that of present-day genotypes: the heaviest casualties were in the Mediterranean, the very regions whose descendents account for the lowest incidences of the HIV-1 resistant allele” (Cohn, QJM, The Black Death and AIDS: CCR5-Delta32 in genetics and history). There are MANY other treatments of the topic. There are many hypotheses on the matter that warrant further research. The bubonic plague hypothesis just isn’t one of them.

I live with HIV. My diagnosis 18 years ago was no problem because I had always educated myself about the virus. The reality I never expected was how disrespectful people would become. My own family put me in a hotel room that they purchased for exactly 7 days. The last words spoken to me were, "_call us once or twice a year and let us know you're still alive_". The woman who raised me as one of her own children is the person who spoke those words to me as she hugged me and then let go of me to get back in her car and drive away. To this day, after I made numerous attempts to redeem myself with my family, they still don't want me around. In closing. When I hear these stories of people being cured of HIV, I am happy for them because they survived. 🙏

Wasn’t there a doctor in China that used CRISPR to remove the HIV receptors from unborn twins with a positive mother? If stem cells can be extracted from bone marrow; why don’t they just use Crispr to modify the patients stem cells? Wouldn’t that prevent rejection?
