Lansky didn’t know what exactly caused the puncture. He just assumed it was a nail. As for why he has his gun out; when you’re illegally delivering a large shipment of liquor during a time when gangs are fighting each other, you take precautions. Especially when you’re having to suddenly stop in the middle of an isolated country road at night. Lansky pulled out his gun as a precaution, turns out he was right - it was an ambush.
