This method is used for analysis of proteins, not DNA. We need to charge the polypeptide chains using SDS. Thus all of them become negatively charged to be movable in the electric field.

No, the glycerol, which has been added to each sample, increases the solution density, thus allowing the sample to sink to the bottom of the well. It is not necessary to add more glycerol on the top of the well.
