Could the world runes be an explanation for some of the unknown events in Runeterra? Fizz's home going missing, Nautilus's transformation in the depths? These runes could provide with a lot more unexplained lore on different champions.

I liked this new lore, as it didn't change the original lore too much, it focused on Ryze's more prominent part (the scrol) and even expanded on his own goal. Not to mention that we now know how did the Void managed to set itself on Icatia. More changes like this and no more like Pantheon's.
When I heard his new voice I thought they changed him to a very generic hero, but after listening to this, like wow, thats pretty dramatic ._. So glad they did some good justice for one of their poster boys haha

I think the reason the runes make people like that is like the lore with Thresh. Some of the spells in the vault actually called to people to use them, and he would deny them the cast by muffling out the last part of the spell.
at 4:30 Icathia has fallen right? well Icathia neighbors the void so maybe that world rune unleashed there could have created the void and the hideous creatures that come from it such as: Kha'zix, Rek'Sai, and finally Kog'Maw

The runes probably give you the power to manipulate the palnet however you want (at least a part of it with one rune). I would like to say that they let you manipulate all matter or space but for runes that are supposed responsible for only creating this one world that would be a bit much (unless it woudl be limited).

The world rune of Icathia could've created a void portal or at least created the potential by perhaps weakening the barriers between worlds/dimensions in some way. As Kassadin and Malzahar both came into contact with the void it is often speculated that other void born came through there such as Vel'koz, Cho'gath, and Kog'maw. While there is no direct evidence to say there are world runes in the shadow isles, Hecarums lore states powerful magical artifacts caused a chain reactive event using a spell to revive the dead amplifying its effects its effects many times over. A world rune would of course be considered a very powerful artifact.
