Sickness is caused by a Pal being low Hunger and Sanity for a long time. This usually happens if a Pal's pathfinding gets stuck and he's unable to get to your feedbox or hotsprings to recover hunger and sanity. You can reset a Pal's position by deposting him in the base and dropping them back out and they'll usually immediately run towards the feedbox. If you're playing in a dedicated server, your base pals will sometimes stop moving if you're logged off preventing them from eating. It's usually better to deposit all your base pals when logging off if you keep logging in to sick Pals.
Medicine takes awhile to craft, the low and mid medi supplies can be bought with gold from the pal merchants like the small pal settlement near the kingpaca boss to the left of the red autumn mountain area/west of the rayne syndicate tower. You can buy high grade, but its way more costly at 3k each. Alternatively then finding your pal and chasing them like trying to give real children shots, you can put them in your team from the pal box and use the medicine from your inventory, it wont let you use the wrong medicine either so if it can be used to cure a pal in your party, you can see and select which to use it on.
Also, a solid lv 3 medicine maker that does nothing else at mid 20s, is Felbat found in a cave north of the elizabee world boss. Alternatively, you can breed one if you have a spare cake by breeding a Penking with either a Chikipi or a teafant
