    I have a few questions as stated below:
    1) why did we attach the negative electrode to the SDS-gel if it already contains a negative component i.e. the sulphate that helped to denature the protein?
    2) If multiple Ab binds to a specific protein,is there a chance to get improper result?
    3) When did we add the Luminol?

      A key part of the lecture he missed is the role of SDS and that West blotting detects Amino acid (AA) sequence for proteins (other blots detect nucleic acid sequence).
      AA have a Verity of charges. On treating the denatured protein (I.e AAs) with SDS, the SDS coats the surface of the unfolded AAs to give the protein a net negative charge. Hence the proteins have a -ve charge and move towards the positive electrode attached to the membrane. You need both -ve and +be fields to run electrophoresis.
      2. Antibodies are usually highly specific to antigens, but in this case,some stray away and bind to the membrane (Hence "blocking" is necessary or you get false +ve results)
      3. The secondary antibody which binds to the primary antibody (an anti-immunoglobulin) is conjugated with an enzyme eg Horse radish peroxidase (HRP).
      HRP catalyzes the degradation of the substrate luminol. As Luminol degrades, it emits light the position of which is detectable on x-ray film

I have only very simple note about this video .. dear brother when you draw the membrane after blotting , the transferred bands should be resemble to bands you did draw on gel after electropheresis , bcz this step only for transferring , but at the last step ( probing ) this will not happen bcz only target protein will appear .     Sir you said when current passed from gel (-ve charged) to membrane (+ve charged) by by conventional we know current move from " +ve to -ve". Then how this imprinting is formed? And why the gel has to be negatively charged?
