Prof, i would like to ask, if i have 2 different techniques to compare with (before and after treatments), and they are 2 different subjects to evalute the effectiveness of the techniques is it possible for me to use independent samples t-test? Thanks.

so what if it is not significant at 2=tailed also which the result will be insignificant?
Yes, if two-tailed test is insignificant, the hypothesis is not supported.
If our total sample size is say 200, out of which 145 males and only 55 females. Here is asymmetry between the two groups, so is there any problem in applying the t test, sir?
Yes, this could be a problem. Better you go for Mann Whitney U Test.
The difference is sample size is quite large
