Good Morning. I would like to know, what are the reagents that were used to create the Pikovskaya culture medium? Because I tried it with versions of PDF documents, but I did not get good results (all the boxes are contaminated)
Hi sajid i m trying to make a culture of psb by extracting it from solid psb biofertilizer product which we bought from the market, after making suitable medium of 500 ml n aft sterlization...we transfer the solution of psb product mixed with distill water..in the medium and then we keep the flask of medium n bacteria on shaker daily for around 4..5 hrs.. but we still cant see proper growth of bacteria...can u help or suggest something..thanx in advace
hi Nayan.. I hope you are using pikovskaya media for the same. make sure the media is made properly. many time bacteria grow slowly on this media. I suggest you to isolate them on nutrient agar with some glucose and then transfer on pikovskaya agar. use saline instead of distilled water for making suspension. pikovskaya should be white in colour, if not.. something went wrong..
I've tried the surface level soil around a healthy growing plant..no results yet...should the soil be taken from agricultural field,?..which is the best one pls suggest

I've tried with PVK..but not the himedia...i weighed the components individulally.
hi laxmi.. chances of isolation increases with number of different samples. personally I found rhizosphere sample the best

How to calculate solubilization index for the colony showing a halo zone..because, so far I just did a single streak of the isolate..
Ratio of total diameter (colony + zone) and colony diameter × 100
Sajid Shaikh okay..but for that should be put a certain micro liters and then check for zone diameter?..
lakshmi sruthi not necessary... just spot inoculate.... liquid media will give more correct estimate of solubization....

yes but it also depends upon type of sample
