Do you have the original version of spire or a cracked one? If you have a cracked one I think there might be an issue then. If you have the original one then try to turn the cutoff wheel in spire and then go to tools and create a automation clip.
It depends ob which genre you want to make. If Trance I would recommend Spire - Diva - Serum. I can't say anything about Plex because I have never worked with it

Probably could improve the pluck by adding a bit of overdrive to filter, or by introducing a "pitch envelope" like in Ian Betts leads and like in a preset "Elegance" from Synthation presets bank for V-Station VSTi.
