I have a request, plz guide, can we automatically measure cross-sectional area for different colored particles in one image in a way it can detect, measure and enlist all the particles i.e. their number, their respective areas instead of considering them same kind of particles?

Abdul Salam you can use an image measured from the same camera and magnification to set a scale. In biology people often use hemocytometer to capture an image and set a scale

Why did we have to change to type to 8-bit for measuring?
Perhaps it is easier to process the images with this configuration, in addition to its lower weight.

You ae right. The threshold need not be the same in every sample.

I have question ...if our image has no scale it's taken from smart phone then how it's work ?
For making length and area measurements, a scale is mandatorily needed with the image.
anyone feels like image-j calculates porosity lower than it actually is? (I am calculating porosity of epoxy impregnated rock thin sections)

SMS, we need to determine the area of a JPEG image and then within that area an anomaly.... that way we can compute the percent that the anomaly is of the total area. Do you recommend ImageJ or jiji ImageJ? We cant calibrate or determine the area of size within the image, that is why we need to determine the percent. The 1st area is essentially a circle the 2nd inside area is somewhat of a triangle. Thanks in advance.

Is there anyway to automate finding distance between points? I am trying to find the distance between neighbor atoms in a lattice from a TEM image but it would be cumbersome to do it one by one.

That is the scale bar which you have to obtain directly from microscope using any image capturing software like LEICA IMAGE ANALYSER.

Hi professor. Great video! I will appreciate so much if you can clarify my doubt, it is about the result of the mean value obtained by the thresholding method. The summarized results showed 2851.485nm^2 +/- 1217nm^2. So, is it possible to say that the grain size of your sample is: 2851.485^0.5 +1217^0.5 = 53.40 nm +/- 34.89 nm ?? Thanks for your time
that would obviously depend on the shape you want to assume. Most particles shapes are approached using spheres so you would need to find the diameter of a sphere with given cross sectional area.. if the shape is very irregular i would either take an average of different lines through the particle or choose a strategy (like e.g. finding the maximum Feret diameter)

Why do we need to convert the image to an 8-bit mode? Is there any specific reason for this?
conversion to 8-bit improves the quantification because it converts the pixel into binary i.e white or black

There is no area. It is a linear distance.

I was wondering how can use the threshold if I have holes onto a surface area that I want to measure? It feels like they break my total area in small pieces. I was thinking to take about 5 measurement of it manually at this point and use a standard deviation, could that work?
Yes. It should work perfectly.
When I scanned my leaf, I forgot to put ruler beside it so I cannot do set scale. what do you think I should do?
If you know the length of any part of the leaf ( which you can measure manually after scanning using a scale), just draw a straight line over this know length. Using set scale, give it the value, which is measure manually. This process will not be 100% accurate, but it will work.

That is the scale of the image which you can obtain directly from microscopy.

Yes. With the freehand option in the task bar you can draw the silhouette of the particle

Is there a way to measure the selected rectangular area so that the same area of interest can be kept uniform for multiple images? or if we select the rectangular area in one image and are analyzing a stack of images, will that same rectangular area selection will apply to all?
